Veel organen zijn afhankelijk van stamcellen voor hun ontwikkeling tijdens de embryogenese en voor onderhoud of reparatie tijdens het volwassen leven. Te begrijpen hoe stamcellen vormen, en hoe ze omgaan met hun omgeving is daarom van cruciaal belang voor het begrijpen van de ontwikkeling, homeostase en ziekte. De eierstok van de fruitvlieg Drosophila melanogaster heeft gediend als een invloedrijk model voor de interactie van kiembaan stamcellen (GSCs) met hun somatische ondersteuning cellen (niche) 1, 2. De bekende locatie van de niche en de GSCs, gekoppeld aan de mogelijkheid om genetisch manipuleren, heeft het mogelijk onderzoekers een groot aantal interacties tussen stamcellen en hun niches 3-12 toe te lichten. Ondanks de overvloed aan informatie over de mechanismen die de SGR onderhoud en differentiatie, relatief weinig bekend over hoe GSCs en hun somatische niches vormen tijdens de ontwikkeling. Ongeveer 18 somatische niches, waarvan cellulaire componenten omvatten terminal filament en cap cellen (figuur 1), vormen tijdens het derde larvale instar 13-17. GSCs afkomstig uit primordiale kiemcellen (PGC). PGC&#39;s verspreiden zich in de vroege larvale stadia, maar na de vorming van de niche een subgroep van PGC&#39;s wordt GSCs 7, 16, 18, ​​19. Samen vormen de somatische niche cellen en de GSCs maken een functionele eenheid dat eieren produceert gedurende de hele levensduur van het organisme. Veel vragen met betrekking tot de vorming van de SGR-eenheid blijven onbeantwoord. Processen, zoals de coördinatie tussen de voorloper cellen voor niches en stamcellen voorlopers, of het genereren van asymmetrie in PGC&#39;s naarmate ze GSCs, kan het beste worden bestudeerd in de larve. Echter, een methodische studie van de larvale ontwikkeling ovarium is fysiek uitdagend. Ten eerste larvale eierstokken zijn klein. Zelfs op late larvale stadia zij slechts 100 urn over te brengen. Wij bieden een aantal technische oplossingen voor problemen zoals het lokaliseren van de eierstokken, vlekken en het wassen van weefsels die niet zinken, en ervoor te zorgen dat de antilichamen doordringen in het weefsel. Dit protocol kan worden toegepast op eerdere larvale stadia en larven testes ook.
