Whole-mount immunohistochemische analyse voor de beeldvorming van de hele vasculatuur is cruciaal voor het begrijpen van de cellulaire mechanismen van vertakking morfogenese. We hebben de ledemaat huid vasculatuur model vasculaire ontwikkeling, waarbij een reeds bestaande primitieve capillaire plexus is omgevormd tot een hiërarchisch vertakte vasculaire netwerk te bestuderen. Whole-mount confocale microscopie met meerdere etikettering maakt voor robuuste beeldvorming van intacte bloedvaten evenals hun cellulaire componenten, waaronder endotheelcellen, pericyten en gladde spiercellen, met behulp van specifieke fluorescente merkers. Vooruitgang in dit onderdeel de huid vaatstelsel model met genetische studies hebben een beter begrip moleculaire mechanismen van vasculaire ontwikkeling en patroonvorming. De ledematen huid vasculatuur model is gebruikt om te bestuderen hoe perifere zenuwen een ruimtelijk model voor de differentiatie en patroonvorming van de slagaders. Deze video artikel beschrijft een eenvoudige en robuuste protocol om intact bloedvaten vlek met vasculaire specifieke antilichamen en fluorescente secundaire antilichamen, die van toepassing is voor de gevasculariseerd embryonale organen waar we in staat zijn om het proces van vasculaire ontwikkeling te volgen.
