Zwei Elektroden Voltage-Clamp-Elektrophysiologie (TEVC) ist ein leistungsfähiges Werkzeug, um den Mechanismus von Ionen-Transport1 für eine Vielzahl von Membranproteinen einschließlich Ionenkanäle 2, Ionenpumpen 3 und Transporter 4 Untersuchen. Dies wird durch selektives Anbringen Donor und Akzeptor Fluorophore zu zwei mutierte Cystein-Reste von Interesse erzielt. In Kürze sind diese Versuche folgende funktionelle Expression des gewünschten Proteins auf der Oberfläche des Xenopus-Oozyten leavis durchgeführt. Die große Oberfläche dieser Eizellen ermöglicht einfache funktionelle Messungen und eine robuste Fluoreszenzsignal 5. Es ist auch möglich, ohne Änderung der extrazellulären Bedingungen wie pH, Liganden oder Kationen / Anionen, die weitere Informationen über den Mechanismus von Membranproteinen 4 kann liefern. Schließlich haben die jüngsten Entwicklungen auch die Manipulation der Auswahl der internen Ionen folgenden Co-Expression mit einem zweiten Protein 9 aktiviert. Unser Protokoll ist in mehreren Teilen beschrieben. Zunächst ging Cystein-Scanning-Mutagenese von Fluorophor Kennzeichnung auf Rückstände an der Schnittstelle der Transmembran-und extrazellulären Domänen befindet sich abgeschlossen ist.
