chromatin immunoprecipitation assay for tissue-specific genes using early-stage mouse embryos

Chromatin-Immunopräzipitation (ChIP) ist ein leistungsfähiges Werkzeug, um Protein zu identifizieren: Chromatin-Wechselwirkungen, die im Kontext von lebenden Zellen 1-3 auftreten. Diese Technik wurde in großem Umfang in Gewebekulturzellen ausgebeutet und in geringerem Maße, in primärem Gewebe. Die Anwendung des ChIP mit Nagetier-embryonalen Gewebe, insbesondere in den frühen Zeiten der Entwicklung ist durch die begrenzte Menge an Gewebe und die Heterogenität der Zell-und Gewebetypen des Embryos kompliziert. Hier präsentieren wir eine Methode, um ChIP durchführen mit einem dissoziierten Embryonaltag 8,5 (E8.5) Embryo. Geschert Chromatin aus einem einzigen Embryo E8.5 können unterteilt werden in bis zu fünf Portionen, die die Ermittler genügend Material ermöglicht Kontrollen und für die Untersuchung von spezifischen Protein: Chromatin-Wechselwirkungen. Zunächst Immunpräzipitation von gewebespezifischen Faktoren nicht unbedingt isoliert Chromatin aus dem Zelltyp, wo der Faktor ausgedrückt. Zweitens sollte Immunpräzipitation von Ko-Aktivatoren und Histonen mit post-translationale Modifikationen, die mit Gen-Aktivierung assoziiert sind nur bei Genen und Genprodukten regulatorische Sequenzen in die Zelle Typ, bei dem das Gen wird gefunden werden oder wurde aktiviert.
