Bakgrund: Tidigare visade vi att neutraliserande förmåga, men inte den koncentration av GM-CSF autoantikroppar var korrelerad med sjukdomens svårighetsgrad hos patienter med autoimmun pulmonell alveolär proteinosis (PAP) 1-3. Eftersom upphävandet av GM-CSF bioaktivitet i lungan är den troliga orsaken till autoimmuna PAP 4,5, det är lovande att mäta neutraliserande förmåga GM-CSF autoantikroppar för utvärdering av sjukdomens svårighetsgrad hos varje patient med PAP. Hittills har neutraliserande förmåga GM-CSF autoantikroppar bedömts genom att utvärdera tillväxthämning av mänskliga benmärgsceller eller TF-1 celler stimuleras med GM-CSF 6-8. I bioassay systemet, dock är det ofta problematiskt att få tillförlitliga data samt att jämföra data från olika laboratorier, på grund av tekniska svårigheter att upprätthålla celler i ett konstant tillstånd. Syfte: Att imitera GM-CSF bindning till GM-CSF receptor på cellens yta med hjälp av cellfria receptor-bindande-analys. METODER: Transgenic silkesodlingssektorn teknik har tillämpats för att få en stor summa för rekombinanta lösliga GM-CSF receptor alfa (sGMRα) med hög renhet 9-13. Den rekombinanta sGMRα fanns i den hydrofila sericin lager av siden trådar utan att bli smält i siden proteiner, och därmed kan vi enkelt utdrag ur kokonger i god renhet med neutrala vattenlösningar 14,15. Lyckligtvis oligosackarid strukturer, som är avgörande för bindning med GM-CSF, är mer lik den struktur av mänskliga sGMRα än de som produceras av andra insekter eller jäst. RESULTAT: Den cellfria analyssystem med sGMRα gav data med hög plasticitet och tillförlitlighet. GM-CSF bindning till sGMRα var dosberoende hämmas av polyklonal GM-CSF autoantikroppar på ett liknande sätt som det biologiska testet med TF-1 celler, vilket tyder på att vår nya cellfria analyssystem med sGMRα är mer användbart för mätning av neutraliserande aktivitet av GM-CSF autoantikroppar än bioassay systemet med TF-1 celler eller mänskliga benmärgsceller. SLUTSATSER: Vi etablerade en cell utan analysen kvantifiera neutraliserande förmåga GM-CSF autoantikroppar.
