La microscopie cryo-électronique et en trois dimensions de classification et les méthodes présentées ici permettent en moyenne pour la détermination des structures tridimensionnelles de complexes glycoprotéine d&#39;enveloppe à la résolution 20 Å ~. Montage des rayons X des coordonnées des composants monomériques aux cartes de densité permet d&#39;interprétation structurelle au niveau moléculaire. L&#39;amélioration continue dans l&#39;équipement informatique à bas coût combiné avec les développements dans les outils de l&#39;open source scientifique et la prolifération des infrastructures de réseau rendent possible auparavant inimaginables scientifiques efforts de collaboration. Nous profitons de ces développements et de montrer que des techniques avancées scientifiques peuvent être mis à la portée des élèves de plusieurs siècles.

Depuis sa découverte il ya 30 ans, plus de 60 millions de personnes ont été infectées par le virus de l&#39;immunodéficience humaine (VIH) <a href="http://www.usaid.gov">(www.usaid.gov)</a> . Le virus infecte et détruit les cellules CD4 + T-cellules ainsi paralysant le système immunitaire, et provoquer un syndrome d&#39;immunodéficience acquise (SIDA) 2. L&#39;infection débute lorsque la glycoprotéine d&#39;enveloppe du VIH &quot;pic&quot; en contact avec le récepteur CD4 à la surface de la T CD4 + cellule. Cette interaction induit un changement conformationnel dans le pic, ce qui favorise l&#39;interaction avec une surface de la cellule seconde co-récepteur 5,9. L&#39;importance de ces interactions protéine dans la voie d&#39;infection à VIH les rend d&#39;une importance capitale en matière de VIH de la recherche fondamentale, et dans la poursuite d&#39;un vaccin contre le VIH. La nécessité de mieux comprendre les interactions à l&#39;échelle moléculaire de la cellule de contact et de neutralisation du VIH a motivé le développement d&#39;une technique pour déterminer lesstructures de la flambée du VIH interagissant avec des protéines cellulaires de surface des récepteurs et des molécules qui bloquent l&#39;infection. Utiliser microscopie cryo-électronique et traitement d&#39;images 3D, nous avons récemment démontré la capacité de déterminer de telles structures sur la surface du virus natif, à la résolution 20 Å ~ 9,14. Cette approche n&#39;est pas limitée aux structures résoudre enveloppe du VIH, et peut être étendu à d&#39;autres protéines de la membrane virale et protéines reconstituées sur un liposome. Dans ce protocole, nous décrivons comment obtenir des structures des glycoprotéines d&#39;enveloppe du VIH à partir de virions VIH purifié et de procéder par étapes dans la préparation des échantillons vitrifiés, la collecte, les données de microscopie cryo-électronique, la reconstitution et le traitement 3D des volumes de données, avec une moyenne et la classification des sous-volumes de protéines en 3D, et l&#39;interprétation des résultats à produire un modèle de protéine. Les aspects informatiques de notre approche ont été adaptés en modules qui peut être consulté et exécuté à distance en utilisant l&#39;Biowulf GNU / Linux en parallèle ppôle rocessing au NIH <a href="http://biowulf.nih.gov">(http://biowulf.nih.gov)</a> . Cet accès à distance, combinée avec le matériel informatique à faible coût et d&#39;accès réseau à grande vitesse, a rendu possible la participation de chercheurs et d&#39;étudiants qui travaillent à l&#39;école ou à domicile.
