De procedure van vibratome snijden, gevolgd door immunohistochemie en confocale beeldvorming zorgt voor de visualisatie van het orgaan van Corti met minimale weefselbeschadiging. De zoogdieren orgaan van Corti is een sterk geordende cellulaire mozaïek van mechanosensorische haar en nonsensory ondersteunende cellen (Beoordeeld in 1,2). Visualisatie van dit cellulair mozaïek vereist vaak dat het orgaan van Corti is cross-delig. In het bijzonder kan de nonsensory pijler en Deiters &#39;cellen, waarvan de kernen zijn basaal gelegen met betrekking tot de haarcellen, niet zichtbaar worden zonder cross-opdeling het orgaan van Corti. Echter, de delicate cytoarchitecture van de zoogdieren orgaan van Corti, met inbegrip van de fijne cytoplasmatische processen van de pijler en Deiters &#39;cellen, is moeilijk te behouden door routine histologische procedures, zoals paraffine en cryo-snijden, die compatibel zijn met standaard immunohistochemische kleuring technieken. Hier beschrijf ik een eenvoudige en deugdelijke procedure die bestaat uit vibratome opdeling van het slakkenhuis, immunohistochemische kleuring van deze vibratome secties in hele berg, gevolgd door confocale microscopie. Deze procedure is op grote schaal gebruikt voor immunhistochemical analyse van veelvoudige organen, waaronder de muis ledematen knop, zebravis darmen, lever, pancreas en hart (zie 3-6 voor geselecteerde voorbeelden). Daarnaast is deze procedure was succesvol voor zowel beeld-als quantitificaton van de zuil aantal cellen in de mutant en controle organen van Corti in zowel embryo&#39;s en volwassen muizen 7. Deze methode is echter momenteel niet op grote schaal gebruikt om de zoogdieren orgaan van Corti te onderzoeken. Het potentieel voor deze procedure tot een betere bescherming bieden beide van de boete cytoarchitecture van de volwassen orgaan van Corti en zorgen voor kwantificering van de verschillende celtypen wordt beschreven.
