Tijdens de metastase verspreiden kankercellen uit de primaire tumor, binnen te dringen in de omliggende weefsels, en de verspreiding naar verre organen. Metastase is een complex proces dat kan gaan om vele soorten weefsel, overspanning variabele periodes en zich vaak diep in organen, waardoor het moeilijk te onderzoeken en te kwantificeren. Bovendien is de effectiviteit van de metastatische proces beïnvloed door de verschillende stappen in de metastatische cascade waardoor het moeilijk is om de bijdrage van een enkel aspect van de tumor cel gedrag te evalueren. Als gevolg hiervan zijn metastase testen vaak uitgevoerd in proefdieren om een ​​realistische context noodzakelijk om in te studeren metastase te bieden. Helaas zijn deze modellen nog verder gecompliceerd door hun complexe fysiologie. Het kuiken embryo is een uniek in vivo model dat veel beperkingen overwint aan het bestuderen van metastase, te wijten aan de bereikbaarheid van de chorion-membraan (CAM), een goed gevasculariseerd extra-embryonaal weefsel bevindt zich onder de eierschaal die ontvankelijk is voor de xenografting van tumorcellen (figuur 1). Aangezien het kuiken embryo is natuurlijk immunodeficiënte, de CAM ondersteunt gemakkelijk de innesteling van zowel de normale en tumor weefsel. Het belangrijkste is dat het aviaire CAM ondersteunt met succes de meeste kankercellen kenmerken, zoals groei, invasie, angiogenese, en herinrichting van de micro-omgeving. Dit maakt het model bijzonder nuttig voor het onderzoek van de routes die leiden tot kanker metastase en de reactie van uitgezaaide kanker aan nieuwe potentiële geneesmiddelen te voorspellen. De detectie van verspreide cellen door soortspecifieke Alu PCR maakt het mogelijk om kwantitatief te beoordelen uitzaaiingen in organen die zijn gekoloniseerd door zo weinig als 25 cellen. Met behulp van de Human epidermoïde carcinoom cellijn (HEp3) gebruiken we dit model tot spontane uitzaaiing van kankercellen te analyseren naar verre organen, waaronder het kuiken lever en longen. Bovendien, met behulp van de Alu-PCR-protocol tonen we aan de gevoeligheid en reproduceerbaarheid van de test als een instrument voor het analyseren en intravasation, arrestatie, extravasatie, en kolonisatie te kwantificeren als individuele elementen van metastase.
