protein membrane overlay assay: a protocol to test interaction between soluble and insoluble proteins in vitro

Valideren van interacties tussen verschillende eiwitten is van vitaal belang voor het onderzoek van hun biologische functies op het moleculaire niveau. Er zijn verschillende methoden, zowel in vitro als in vivo, de eiwitbinding te evalueren, en ten minste twee methoden die de tekortkomingen van elkaar aan te vullen moeten worden uitgevoerd om betrouwbare inzichten te verkrijgen. Voor een in-vivo assay, de bimoleculaire fluorescentie invulling (BiFC) test de meest populaire en minst ingrijpende aanpak die in staat stelt om eiwit-eiwit interacties te detecteren binnen levende cellen vertegenwoordigt, evenals identificeren van de intracellulaire lokalisatie van de interagerende eiwitten 1,2. In deze test, niet-fluorescerende N-en C-terminale helften van GFP of zijn varianten gefuseerd met getest eiwitten, en wanneer de twee fusie-eiwitten bij elkaar worden gebracht als gevolg van de geteste eiwitten &#39;interacties, is het fluorescerende signaal gereconstitueerde 3-6 .
