mapping inhibitory neuronal circuits by laser scanning photostimulation

Remmende neuronen zijn cruciaal voor corticale functies. Zij omvatten ongeveer 20% van de gehele corticale neuronale populatie en kan verder worden onderverdeeld in verschillende subtypes gebaseerd op hun immunochemische, morfologische en fysiologische eigenschappen 1-4. Hoewel eerder onderzoek heeft veel onthuld over de intrinsieke eigenschappen van afzonderlijke soorten remmende neuronen, kennis over hun lokale circuit verbindingen is nog relatief beperkt 3,5,6. Gezien het feit dat elk individueel neuron de functie wordt gevormd door de exciterende en inhiberende synaptische input binnen corticale circuits, hebben we met behulp van laser scanning fotostimulatie (WAPZ) naar de kaart lokale circuit verbindingen met specifieke remmende celtypen. Vergeleken met conventionele elektrische stimulatie of glutamaat bladerdeeg stimulatie, WAPZ heeft unieke voordelen waardoor uitgebreide mapping en kwantitatieve analyse van de lokale functionele ingangen individueel geregistreerd neuronen 3,7-9. Laser fotostimulatie via de glutamaat uncaging activeert selectief neuronen perisomatically, zonder activering van axonen van doorgang of distale dendrieten, die een sub-laminaire mapping resolutie zorgt ervoor. De gevoeligheid en efficiëntie van WAPZ voor mapping inputs van vele plaatsen stimulatie over een groot gebied zijn goed geschikt voor corticale circuit analyse. Hier introduceren we de techniek van WAPZ in combinatie met whole-cell patch clamping voor lokale remmende circuit in kaart brengen. Gerichte opnamen van specifieke remmende celtypen vergemakkelijkt door het gebruik van transgene muizen die groen fluorescent eiwit (GFP) in beperkte remmende neuron populaties in de cortex 3,10 Dat sluit bemonstering van de beoogde celtypes en ondubbelzinnige identificatie van de opgenomen celtypen kunnen . Zoals voor WAPZ in kaart brengen, schetsen we het systeem instrumentatie, beschrijven de experimentele procedure en data-acquisitie, en de huidige voorbeelden van circuit in kaart brengen in muis primaire somatosensory cortex. Zoals geïllustreerd in onze experimenten wordt opgesloten glutamaat geactiveerd in een ruimtelijk beperkte gebied van de hersenen slice door UV laser fotolyse, gelijktijdige voltage-clamp opnamen waarmee de aanwezigheid van fotostimulering opgewekte synaptische responsen. Kaarten van zowel stimulerende of remmende synaptische input aan de beoogde neuron worden gegenereerd door het scannen van de laserstraal honderden potentiële presynaptische locaties te stimuleren. Zo WAPZ maakt de bouw van gedetailleerde kaarten van synaptische input botsende op specifieke soorten remmende neuronen door middel van herhaalde experimenten. Tezamen de fotostimulering gebaseerde techniek biedt neurowetenschappers een krachtig hulpmiddel voor het bepalen van de functionele organisatie van de lokale corticale circuits.
