De oprichting van een functioneel bloedvat-netwerk is een essentieel onderdeel van organogenese, en is noodzakelijk voor een optimale orgaanfunctie. Bijvoorbeeld, in de thymus juiste vaatstelsel vorming en patroonvorming is essentieel voor thymocyt toegang tot het orgel en de mature T-cel uitgang naar de periferie. De ruimtelijke ordening van de bloedvaten in de thymus is afhankelijk van signalen uit de lokale micro-omgeving, namelijk de thymus epitheelcellen (TEC). Diverse recente rapporten suggereren dat verstoring van deze signalen leidt tot thymus bloedvat gebreken 1,2. Eerdere studies hebben beschreven technieken die gebruikt worden om de neonatale en volwassen thymus bloedvaten 1,2 label. We zien hier een techniek voor de etikettering de bloedvaten in de embryonale thymus. Deze methode combineert het gebruik van de FITC-dextran of Griffonia (Bandeiraea) simplicifolia Lectin I (GSL 1 - isolectin B 4) gezichts-ader injecties en CD31 antilichaam kleuring om thymus vasculaire structuren en PDGFR-β aan thymus perivasculaire mesenchym 3-5 label te identificeren. De optie van het gebruik van vriescoupes of vibratome secties is ook voorzien. Dit protocol kan worden gebruikt om de thymus vasculaire defecten, wat essentieel is voor het definiëren van de rol van TEC-afgeleide moleculen in thymus bloedvatvorming te identificeren. Als de methode labels van de gehele vaatstelsel, het kan ook gebruikt worden om de vasculaire netwerken te analyseren in meerdere organen en weefsels in het hele embryo inclusief huid en hart 6-10.
