Die Rekrutierung von zirkulierenden Leukozyten aus Blut, um das entzündete Gewebe ist ein wesentlicher und komplexer Prozess der Entzündung 1,2. In den postkapillären Venolen des entzündeten Gewebes, Leukozyten zunächst Leine und rollen auf der luminalen Oberfläche von venulären Wand. Intravitalmikroskopie ist ein leistungsstarkes Tool zur Visualisierung von Leukozyten-Endothelzell-Interaktionen in vivo und aufschlussreich zellulären und molekularen Mechanismen der Rekrutierung von Leukozyten 2,5. In diesem Bericht stellen wir eine umfassende Beschreibung der Verwendung von Hellfeld Intravitalmikroskopie zu visualisieren und zu bestimmen, die genauen Abläufe des neutrophilen Rekrutierung in Maus M. cremaster in Reaktion auf den Verlauf eines Neutrophilen Lockstoff. Um neutrophilen Rekrutierung, ein kleines Stück Agarosegel (~ 1-mm Größe 3) mit neutrophilen chemoattractant MIP-2 (CXCL2, ein CXC-Chemokin) oder WKYMVm (Trp-Lys-Tyr-Val-D-Met, ein synthetisches Analogon zu induzieren von bakteriellen Peptid) wird auf das Muskelgewebe neben den beobachteten postkapillären venule platziert. Mit Zeitraffer-Video Fotografie und Computer-Software ImageJ, Neutrophilen intraluminale Kriechen auf Endothel, Neutrophilen transendotheliale Migration und der Migration und Chemotaxis in Gewebe sichtbar gemacht und verfolgt werden. Dieses Protokoll ermöglicht die zuverlässige und quantitative Analyse von vielen Neutrophilen Rekrutierung Parameter wie intraluminale krabbeln Geschwindigkeit, Seelenwanderung Zeit, Distanz Zeit, Wanderungsgeschwindigkeit, Chemotaxis Geschwindigkeit und Chemotaxis Index im Gewebe. Wir zeigen, dass dieses Protokoll verwenden, diese neutrophilen Rekrutierung Parameter stabil bestimmt werden können und die einzelne Zelle Fortbewegung bequem in vivo verfolgt.
