Det er allment akseptert at endringer i vaskulær skjærspenning utløse uttrykk av inflammatoriske gener i endotelceller og dermed indusere arteriosklerose (anmeldt i 1 og 2). Rollen skjærspenning har blitt grundig studert in vitro undersøke påvirkning av flyten dynamikk på dyrkede endotelceller 1,3,4 og in vivo i større dyr og mennesker 1,5,6,7,8. Men svært reproduserbare små dyremodeller gir systematisk undersøkelse av påvirkning av skjærspenning på plaque utvikling er sjeldne. Nylig introduserte Nam et al. Ni en mus modell der ligation av grenene av carotisar skaper en region med lav og oscillasjon flyt. Selv om denne modellen fører til endotelial dysfunksjon og rask dannelse av aterosklerotiske lesjoner i hyperlipidemic mus, kan det ikke utelukkes at den observerte inflammatorisk respons er, i hvert fall delvis en konsekvens of endotelceller og / eller fartøy skade på grunn ligation. Deres fenotypen er korrelert med regional flow dynamikken 11 som bekreftes av in vivo Magnetic Resonance Imaging (MRI) 12: Lav og laminær skjærspenning oppstrøms mansjetten fører til dannelse av store plaques i en mer sårbar fenotype, mens oscillasjon skjærspenning nedstrøms mansjetten induserer stabile aterosklerotiske lesjoner 11. I de regionene med høy skjærspenning og høy laminær flow innenfor mansjetten,vanligvis ingen aterosklerotisk plakk er observert. I konklusjonen, er den skjærspenning-endring cuff prosedyre en pålitelig kirurgisk tilnærming til å produsere fenotypisk forskjellige aterosklerotiske lesjoner i APOE-mangelfull mus.
