Dolore neuropatico cronico, derivante da danni al sistema nervoso centrale o periferico, è una condizione diffusa e invalidante, che colpisce il 7-18% della popolazione di 1,2. I sintomi includono spontanea (formicolio, bruciore, scosse elettriche-like) dolore, disestesia, parestesia, allodinia (dolore derivante dalla normalmente non dolorosi stimoli) e di iperalgesia (un aumento della risposta agli stimoli dolorosi). I sintomi sensoriali sono co-morbidità con disabilità comportamentali, come l&#39;insonnia e la depressione. Per studiare il dolore neuropatico cronico diversi modelli animali che mimano lesione del nervo periferico sono stati sviluppati, uno dei più largamente usato è Bennett e Xie (1988) unilaterale nervo sciatico lesione cronica costrizione (CCI) 3 (Figura 1). Qui vi presentiamo un metodo per l&#39;esecuzione di CCI e ipersensibilità al dolore test. CCI viene eseguito in anestesia, con il nervo sciatico da un lato esposto da fare comeincisione parenti, e tagliando il tessuto connettivo tra le superficiale dei muscoli glutei e bicipiti femorale. Quattro legature gut cromico sono legati liberamente intorno al nervo sciatico a intervalli di 1 mm, per occludere solo, ma non arrestare il flusso di sangue epineurale. La ferita viene chiusa con suture nel muscolo e graffe nella pelle. L&#39;animale viene poi lasciato riprendersi dalla chirurgia per 24 ore prima del test ipersensibilità dolore comincia. La soglia di ritiro meccanica è la pressione massima esercitata(In grammi) che fa scattare il ritiro zampa. Per la misurazione di latenza ritiro termico, descritto inizialmente da Hargreaves et al (1988), la zampa posteriore è esposto ad un fascio di calore radiante attraverso una superficie di vetro trasparente usando un misuratore plantare analgesia 5,6. La latenza ritiro allo stimolo di calore viene registrato come il tempo per il ritiro zampa in entrambi i hindpaws feriti e illeso. Dopo CCI, soglia di ritiro meccanica, nonché latenza ritiro termico nella zampa feriti sono entrambi significativamente ridotto rispetto alla misurazioni e la zampa indenne (Figura 3).
