Glioblastoma multiforme (GBM) zijn de meest voorkomende en agressieve volwassen primaire hersentumoren 1. In de afgelopen jaren is er aanzienlijke vooruitgang geboekt in het begrijpen van de mechanismen van tumor invasie en directe intracerebrale inoculatie van de tumor biedt de mogelijkheid van het observeren van de invasieve proces in een fysiologisch geschikte omgeving 2. Voor zover de menselijke hersentumoren betreft, worden de orthotope modellen die nu beschikbaar is tot stand gebracht door stereotaxische injectie van celsuspensies of implantatie van een massief stuk van de tumor door middel van een ingewikkelde craniotomie procedure 3. In de techniek wordt oogsten cellen van weefselkweek aan een celsuspensie gebruikt om rechtstreeks implanteren in de hersenen te maken. De duur van de behandeling is ongeveer 30 minuten, en de muis moet in een constante chirurgische vlak een injecteerbare verdoving wordt gebruikt. De muis is geplaatst in een stereotaxisch mal van Stoetling (figuur 1). Aftre het operatiegebied wordt gereinigd en wordt een incisie gemaakt, en de bregma bevindt zich de plaats van de craniotomie bepalen. De locatie van de craniotomie 2 mm naar rechts en 1 mm rostraal de bregma. De diepte van 3 mm van het oppervlak van de schedel en de cellen worden geïnjecteerd met een snelheid van 2 pi elke 2 minuten.
