I. Glioblastoma multiforme (GBM) ist die häufigste und aggressivste erwachsenen primären Hirntumoren ein. In den letzten Jahren hat es erhebliche Fortschritte im Verständnis von den Mechanismen der Tumorinvasion und direkter intrazerebraler Inokulation des Tumors bietet die Möglichkeit der Beobachtung der invasiven Verfahren in einem physiologisch geeigneten Umgebung 2. Soweit menschlichen Hirntumoren betrifft, so sind die derzeit verfügbaren Modelle orthotopen entweder durch stereotaktische Injektion von Zellsuspensionen oder Implantation eines festen Tumors Stück durch ein kompliziertes Verfahren Kraniotomie 3 hergestellt. In unserer Technik ernten Zellen aus Gewebekulturen, um eine Zellsuspension verwendet, um direkt in das Gehirn implantiert zu schaffen. Die Dauer der Operation beträgt etwa 30 Minuten, und die Maus muss in einem konstanten chirurgischen Ebene liegen, eine injizierbare Anästhetika. Die Maus wird in einen stereotaktischen Bohrlehre von Stoetling (Abbildung 1) aus angeordnet. Achterner das Operationsgebiet wird gereinigt und vorbereitet, wird ein Einschnitt vorgenommen, und die Bregma befindet, um die Position der Kraniotomie bestimmen. Die Lage der Kraniotomie ist 2 mm nach rechts und 1 mm rostral zu Bregma. Die Tiefe von 3 mm von der Oberfläche des Schädels und die Zellen werden mit einer Rate von 2 ul alle 2 Minuten injiziert. Die Haut wird mit 5-0 PDS genäht, und die Maus darf aufwachen auf einem Heizkissen. Aus unserer Erfahrung, abhängig von der Zell-Linie, kann die Behandlung Platz 7 bis 10 Tagen nach der Operation. Drug Delivery ist abhängig von der Arzneimittel-Zusammensetzung. Für Bestrahlung die Mäuse sind anästhesiert, und in einem speziell angefertigten Vorrichtungsbau. Blei deckt der Maus Körpers und setzt nur das Gehirn der Maus. Das Studium der Tumorentstehung und die Evaluierung neuer Therapien für GBM erfordern genaue und reproduzierbare Hirntumor Tiermodellen. So verwenden wir diese orthotopen Gehirn Modell, um die Interaktion der Mikroumgebung des Gehirns und des Tumors zu studieren, um das zu testen aftlichess von verschiedenen therapeutischen Wirkstoffen mit und ohne Bestrahlung.
