Ondartede hjernesvulster, slik som glioblastoma multiforme, representerer de vanligste primære hjernesvulster og har en dyster prognose fire. Glioblastoma multiforme (GBM) er den vanligste og aggressive voksne primære hjernesvulster en. I de senere årene har det vært betydelig fremgang i forståelsen av mekanikken i tumor invasjon, og direkte intracerebral inokulering av svulst gir muligheten til å observere den invasive prosessen i en fysiologisk riktig miljø 2. Såvidt menneskelige hjernesvulster er bekymret, er orthotopic modellene tilgjengelige etablert enten ved stereotaxic injeksjon av cellesuspensjoner eller implantasjon av et solid stykke tumor gjennom en komplisert kraniotomi prosedyre tre. I teknikken vår høster vi celler fra vev kultur for å skape en celle suspensjon brukes til å implantere direkte inn i hjernen. Varigheten av operasjonen er ca 30 minutter, og som musen må være i en konstant kirurgisk fly, er en injeksjon bedøvelse som brukes. Musen er plassert i en stereotaxic jigg laget av Stoetling (figur 1). AftER det kirurgiske området er ryddet og klargjort, er et snitt laget, og den bregma befinner å bestemme plasseringen av kraniotomi. Plasseringen av kraniotomi er 2 mm til høyre og 1 mm rostral til bregma. Dybden er 3 mm fra overflaten av skallen, og cellene blir injisert med en hastighet på 2 mL hver 2 minutter. Huden er sutureres med 5-0 PDS, og musen er lov å våkne opp på en varmepute. Fra vår erfaring, avhengig av cellelinje, kan behandlingen skje fra 7-10 dager etter operasjonen. Drug levering er avhengig av stoffet sammensetning. For strålebehandling musene bedøves, og satt i en spesiallaget pilken. Lead dekker musens kropp og eksponerer bare hjernen på mus. Studiet av tumorigenesis og evaluering av nye behandlingsmetoder for GBM krever nøyaktige og reproduserbare hjernesvulst dyremodeller. Derfor bruker vi denne orthotopic hjernen modell for å studere samspillet av mikromiljøet av hjernen og svulsten, for å teste effectivenESS av ulike terapeutiske agenter med og uten stråling.
