Es gibt zwei unterschiedliche Klassen von bildgebenden Photorezeptoren in der Retina von Wirbeltieren: Stäbchen und Zapfen. Stäbchen sind in der Lage, einzelne Photonen des Lichts erkennen, während Kegel kontinuierlich arbeiten unter rasch wechselnden hellen Lichtverhältnissen. Die Absorption von Licht durch Stab-und Kegel-spezifischen visuellen Pigmente in den äußeren Segmenten der Photorezeptoren löst eine Phototransduktionskaskade, die sich letztendlich in der Schließung des zyklisch Nukleotid-gesteuerten Kanäle auf der Plasmamembran und Zelle Hyperpolarisation. Dieses Licht-induzierte Veränderung der Membran aktuellen und potenziellen kann als Photoresponse registriert werden, entweder durch klassische Saugelektrode Aufnahmetechnik 1,2 oder durch Aufnahmen transretinal Elektroretinogramm (ERG) von isolierten Netzhaut mit pharmakologisch blockiert postsynaptische Antwort Komponenten 3-5. Das letztere Verfahren kann Arzneimittel-zugänglich dauerhafte Aufzeichnungen aus der Maus Photorezeptoren und ist besonders nützlich für die Herstellung von stabilen Photomorphosen from die knappen und fragilen Maus Kegel. Im Falle von Kegeln kann solche Experimente sowohl in dunkeladaptierten folgenden Bedingungen und intensive Beleuchtung, die im wesentlichen alle bleicht Sehpigment durchgeführt werden, um den Prozess der Membran Photoempfindlichkeit Rückgewinnung beim Dunkeladaptation 6,7 überwachen. In diesem Video zeigen wir, wie Stab-und M / L-Kegel-driven transretinal Aufnahmen aus dunkeladaptierten Maus Netzhaut führen.
