measuring peptide translocation into large unilamellar vesicles

Er is een actieve interesse in peptiden die gemakkelijk celmembranen passeren zonder de hulp van celmembraan receptoren 1. Veel van deze worden aangeduid als cel-penetrerende peptiden, die vaak bekend om hun potentieel als drug delivery vectoren 1-3. Bovendien is er toenemende belangstelling voor antimicrobiële peptiden die werken via niet-membraan lytische mechanismen 4,5, in het bijzonder degenen die bacteriële membranen kan passeren zonder dat de cel-lysis en doden cellen door te interfereren met intracellulaire processen 6,7. In feite hebben de auteurs in toenemende mate gewezen op de relatie tussen de cel-penetrerende en antimicrobiële peptiden 1,8. Een goed begrip van het proces van het membraan translocatie en de relatie tussen peptide structuur en zijn vermogen om transloceren moeten doeltreffend, reproduceerbare assays voor translocatie. Verschillende groepen hebben voorgesteld methoden om translokatie meten in grote unilamellar lipide bolletjes (LUVS) 9-13. LUVS dienen als nuttige modellen voor bacteriële en eukaryote cel membranen en worden vaak gebruikt in TL-peptide studies 14,15. Hier beschrijven we onze toepassing van de methode voor het eerst ontwikkeld door Matsuzaki en collega&#39;s op antimicrobiële peptiden, zoals magainin en buforin II 16,17 overwegen. Naast het leveren van ons protocol voor deze methode, presenteren we ook een directe benadering van de data-analyse die translocatie kunnen het gebruik van deze assay kwantificeert. De voordelen van deze translocatie test in vergelijking met anderen, dat het de potentie om informatie over de snelheid van de membraan translocatie te bieden en vereist niet dat de toevoeging van een fluorescent label, dat kan veranderen peptide eigenschappen 18, aan tryptofaan-bevattende peptiden is. In het kort wordt translocatie mogelijkheid in lipide vesicles gemeten als functie van de Foster Resonance Energy Transfer (FRET) tussen autochtone tryptofaan residu&#39;s en dansyl fosfatidylethanolamine als eiwitten worden geassocieerd met de externe LUV membraan (figuur 1). Cel-penetrerende peptiden worden gesplitst als ze tegenkomen ongeremd trypsine ingekapseld met de LUVS, wat leidt tot distantiëring van de LUV membraan en een daling van de FRET signaal. De daling van de FRET signaal waargenomen voor een transloceren peptide is aanzienlijk hoger dan die waargenomen voor dezelfde peptide wanneer de LUVS zowel trypsine en trypsine-remmer, of wanneer een peptide dat niet spontaan kruis lipidemembranen is blootgesteld aan trypsine-bevattende LUVS bevatten. Deze verandering in fluorescentie biedt een directe kwantificering van peptide translocatie in de tijd.
