Linker histon H1 bindt aan de nucleosoom kerndeeltje en linker DNA, het vergemakkelijken van het vouwen van chromatine in de hogere orde structuur. H1 is essentieel voor zoogdieren ontwikkeling 1 en regelt genexpressie in vivo 2-4. Onder de sterk geconserveerde histon-eiwitten, de familie van de H1 linker histonen is de meest heterogene groep. Er zijn 11 H1 subtypes in zoogdieren die differentieel gereguleerd gedurende de ontwikkeling en in verschillende celtypen. Deze H1 subtypen behoren 5 somatische H1S (H1a-e), de vervanging H1 0, 4 kiemcel specifieke H1 subtypes, en H1x 5. De aanwezigheid van meerdere H1 subtypes die verschillen in het DNA bindingsaffiniteit en chromatine verdichting mogelijkheid 6-9 biedt een extra niveau van de modulatie van chromatine functie. Zo kwantitatieve analyse van individuele uitdrukking H1 subtypen, beide van mRNA en eiwit, is noodzakelijk voor een beter begrip van de regulering van hogereOm chromatine structuur en functie. Hier beschrijven een aantal assays ontwikkeld voor het analyseren van de expressie van afzonderlijke subtypes H1 (figuur 1). opregulatie van verschillende H1 variant genen wordt gemeten door een aantal zeer gevoelige en kwantitatieve reverse transcriptie-PCR (qRT-PCR) assays, die sneller en nauwkeuriger en vereisen minder monsters opzichte van de alternatieve benadering van Northern blot analyse. In tegenstelling tot andere cellulaire mRNA berichten mRNAs meeste histoon genen waarvan de meerderheid van genen H1, ontbreekt een lange polyA staart, maar bevatten een stam-lusstructuur aan het 3 &#39;onvertaalde gebied (UTR) 10. Daarom worden cDNA&#39;s bereid uit totaal RNA door reverse transcriptie met random primers plaats van oligo-dT primers. Realtime PCR met primers specifiek voor elk H1 subtypes (Tabel 1) zijn uitgevoerd om zeer kwantitatieve meting van mRNA van de afzonderlijke H1 subtype verkrijgens. Expressie van housekeeping genen worden geanalyseerd als controle voor normalisatie. De relatieve overvloed aan eiwitten van elk subtype H1 en de kern histonen wordt verkregen door middel van omgekeerde fase hoge prestatie vloeistofchromatografie (RP-HPLC) analyse van het totaal histonen uit zoogdiercellen 11-13. De HPLC-methode en elutie voorwaarden die hier worden beschreven voor een optimale scheiding van de muis H1 subtypes. Door het kwantificeren van de HPLC-profiel berekenen we het relatieve aantal afzonderlijke H1 subtypes binnen H1 familie en het H1 nucleosoom verhouding in de cellen te bepalen.
