De in situ hybridisatie en colorimetrische kleuringsprotocol presenteerde hier voor de hele-mount muggen weefsels en embryo&#39;s is een nuttige techniek voor de lokalisatie van transcripties in specifieke organen en celtypes. Deze procedures zijn een verbetering ten opzichte van onze eerder gemeld methoden, zowel in het stroomlijnen van uitgebreide wasstappen en het verstrekken van aanvullende technische details en reagens bronnen. Onze ervaring is dat colorimetrische detectie van hybridisatie signalen is superieur in gevoeligheid en de helderheid van hybridisatie-signaal in vergelijking met fluorescentie detectie op basis van regelingen. Bovendien colorimetrische detectie omzeilt kwesties in verband met het signaal discriminatie in embryo&#39;s, die inherent auto-TL. Beperkingen in het opsporen van hybridisatie signalen zich voordoen als een lage-overvloed transcripties zijn gericht en achtergrondkleuring is evident. Het verhogen van de hybridisatietemperatuur van 65 ° C is gebleken verminderen teruggrond hybridisatiesignalen, maar niet voorgesteld vervanging voor het ontwerpen unieke doelwitspecifieke RNA probes. Dit protocol is gebruikt om uit te voeren in situ hybridisatie van hele-mount speekselklieren van Aedes aegypti, en eierstokken en embryo&#39;s van Anopheles gambiae, Anopheles stephensi, Ae. aegypti en Culex quinquefasciatus. Deze methode is ook toepasbaar op andere mug weefsels en vermoedelijk van andere insecten. Daarnaast hebben we samengesteld voor het eerst vergelijkende richtlijnen voor de enscenering van de embryonale ontwikkeling in drie vector muggen, Ae. aegypti, An. gambiae en Cx. fatigans. Zij zijn beschreven voor specifieke stammen van deze drie soorten onder discreet kweekomstandigheden. Het is belangrijk op te merken dat de ontwikkeling tijdsverloop kan variëren voor verschillende stammen van muggensoorten en onder verschillende kweekomstandigheden. Hybridisaties in situ supplement aan de gang zijnde inspanningen om anaLyze de transcriptomen van muggen en andere geleedpotigen, en kan een beter beeld van de regulatie van genexpressie in deze organismen te bieden.

Muggen zijn vectoren voor een gevarieerde set van pathogenen, waaronder arboviruses, protozoaire parasieten en nematoden. Onderzoek van de transcripten en gen toezichthouders die zijn uitgedrukt in weefsels waarin de mug gastheer en pathogeen interactie krijgen en in organen die betrokken zijn bij de voortplanting van groot belang zijn voor strategieën om muggen overgedragen overdracht van de ziekte te verminderen en te verstoren ei-ontwikkeling. Een aantal hulpmiddelen zijn gebruikt om te bestuderen en valideren van de tijdelijke en weefsel-specifieke regulatie van genexpressie. Hier wordt beschreven protocollen die zijn ontwikkeld om ruimtelijke informatie, die inzicht waar specifieke genen tot expressie en producten verbetert accumuleren verkrijgen. De beschreven protocol is gebruikt om uitdrukking te valideren en te bepalen accumulatie patronen van transcripten in weefsels die verband houden met muggen overgedragen pathogeen transmissie, zoals vrouwelijke speekselklieren, en subcellulaire compartimenten van de eierstokken en embryo&#39;s, waarin islaat om muggen voortplanting en ontwikkeling.
