פיתוח ותחזוקה של מערכת החיסון CD4 + ותאי T רגולטוריים (Tregs) לתרום סובלנות היקפי צריכה להישאר הומאוסטזיס חיסוניות עם סכום עצום של אנטיגנים עצמיים ו commensal ב ועל גוף האדם. הפרעות במאזן בין Tregs ו דלקת בתאי T קונבנציונאלי עלול לגרום immunopathology או סרטן. למרות הזרקת הטיפולי של Tregs הוכח להיות יעיל במודלים Murine של 1 קוליטיס, אני סוכרת סוג 2, דלקת פרקים שגרונית שתל מול המחלה המארח, 4 הבדלים בסיסיים כמה אדם לעומת העכבר Treg ביולוגיה 5 יש ובכך מנעה הרבה בשימוש קליני. היעדר מספר מספיק טוהר, יציבות וספציפיות הביות של טיפולי Tregs חייבה פלטפורמה דינמית של התפתחות Treg האנושי שבו כדי לייעל את התנאים להרחבת vivo לשעבר שלהם 6. כאן אנו דescribe שיטה בידול של המושרה Tregs (iTregs) מתורם דם אדם יחיד היקפי אשר ניתן לפרק לארבעה שלבים: בידוד של כדוריות דם היקפיים mononuclear, בחירת המגנטי של CD4 + T תאים, על תרבית תאים במבחנה הקרינה הופעלה התא מיון (FACS) של תת התא טי. מאז החתימה Treg גורם שעתוק forkhead תיבת P3 (FoxP3) הוא גורם ההפעלה הנגרמת שעתוק בבני אדם 7 ולא סמן ייחודי אחר קיים, פאנל קומבינטורית של סמנים יש להשתמש כדי לזהות תאים T עם פעילות מדכא. לאחר שישה ימים בתרבית, תאים במערכת שלנו ניתן התחום לתאי ה-T נאיבי, חולצות זיכרון תאים או iTregs המבוססים על הביטוי היחסי של CD25 ו CD45RA. כמו זיכרון תמים ותאי T שונים יש דרישות דיווח קיטוב ו plasticities 8, לפני מיון של האוכלוסייה התא הראשוני T לתוך CD45RA + ו + תת CD45RO יכול בדואר רגיל לבחון התאמות אלה. עולה בקנה אחד עם אחרים, CD25 שלנו היי CD45RA - iTregs רמות גבוהות של אקספרס FoxP3 9, GITR ו CTLA-4 ו 11 רמות נמוכות של CD127 12. בעקבות FACS של האוכלוסייה בכל התאים כתוצאה ניתן להשתמש assay מדכא אשר מעריכה את היכולת ביחס לעכב את הפצת carboxyfluorescein succinimidyl אסתר (CFSE), שכותרתו עצמיים בתאי T.
