Rechnen im Gehirn beruht auf Neuronen reagiert angemessen auf ihre synaptische Eingänge. Neuronen unterscheiden sich in ihrer Ergänzung und Verteilung von Ionenkanälen, die Membran, wie sie synaptische Eingänge reagieren zu bestimmen. Allerdings ist die Beziehung zwischen diesen zellulären Eigenschaften und neuronale Funktion im lebenden Tier nicht gut verstanden. Ein Ansatz, dieses Problem ist, topographisch organisierten neuronalen Schaltkreise, in denen die Position der einzelnen Neuronen Karten auf Informationen, die sie kodieren, oder Berechnungen von ihnen durchgeführten 1 zu untersuchen. Experimente mit Hilfe dieses Ansatzes lassen Grundsätze für die Abstimmung der synaptischen Antworten zugrunde liegenden Informationen Codierung in sensorischen und kognitiven Schaltungen 2,3. Die topographische Organisation der räumlichen Darstellung entlang der dorso-ventralen Achse des medialen entorhinalen Cortex (MEC) bietet die Möglichkeit, zwischen zellulären Mechanismen und Berechnungen zu schaffen iW ichtig für die räumliche Wahrnehmung. Neurone in Schicht II des MEC Nagetier kodieren Lage mit gitterartigen Brennen Felder 4-6. Für Neuronen in dorsalen Position in der MEC vor der Abstand zwischen den einzelnen Brennen Bereichen, die ein Raster bilden in der Größenordnung von 30 cm, während für Neuronen in zunehmend ventralen Positionen dieser Abstand erhöht, um größer als 1 m. Mehrere Studien haben zellulären Eigenschaften von Neuronen in Schicht II des MEC, dass, wie der Abstand zwischen Gitter Brennen Felder, unterscheiden sich auch je nach dorsal-ventrale Position enthüllt, was darauf hindeutet, dass diese zellulären Eigenschaften wichtig sind für die räumliche Berechnung 2,7-10. Hier beschreiben wir Verfahren für die Vorbereitung und elektrophysiologische Ableitung von Hirnschnitten, die dorsal-ventralen Umfang der MEC ermöglicht Untersuchung der topographischen Organisation der biophysikalischen und anatomischen Eigenschaften von MEC Neuronen aufrecht zu erhalten. Die dorsal-ventrale Position des identifizierten neurons relativ zu anatomischen Landmarken ist schwierig, genau festzustellen mit Protokollen, die horizontale Scheiben von MEC 7,8,11,12 zu verwenden, da es schwierig, Anhaltspunkte für die genaue dorsal-ventrale Lage der Scheibe zu etablieren. Die Verfahren beschreiben wir ermöglichen präzise und einheitliche Messung der Lage der aufgenommenen Zellen entlang der dorso-ventralen Achse des MEC sowie die Visualisierung molekularer Gradienten 2,10. Die Verfahren wurden für die Verwendung mit erwachsenen Mäusen (&gt; 28 Tage) entwickelt worden und haben sich bislang mit Mäusen bis zu 1,5 Jahre alt beschäftigt. Mit Anpassungen konnten sie mit jüngeren Mäusen oder anderen Nagetieren eingesetzt werden. Ein standardisiertes System der Vorbereitung und Messung helfen werden systematische Untersuchung der zellulären und Mikroschaltung Eigenschaften dieser Gegend.
