Expressie van vier transcriptiefactoren herprogrammeert humane fibroblasten met iPSCs. Veel pogingen werden gedaan om de menselijke iPSCs behulp van niet-integratie of niet-genetische benaderingen van klinisch veilige iPSCs genereren. Tot nu toe, deze methoden blijkt extreem lage efficiëntie en de verdere optimalisatie van de reproduceerbaarheid 11-14 te verbeteren. Retro-of lentivirale methoden gemakkelijk worden gebruikt voor het afleiden en toe te passen iPSCs voor menselijke in vitro ziekte-mode...

Menselijke embryonale stamcellen (hESC &#39;s) zijn pluripotent en een onschatbare cellulaire bronnen voor in-vitro-en vaatziekten modellering en regeneratieve geneeskunde 1. Het is eerder aangetoond dat de menselijke somatische cellen kunnen worden geherprogrammeerd om pluripotentie door ectopische expressie van vier transcriptiefactoren (Oct4, Sox2, Klf4 en Myc) en worden geïnduceerde pluripotente stamcellen (iPSCs) 2-4. Net als hESC &#39;s, de menselijke iPSCs zijn pluripotent en een potentiële bron voor autologe cellen. Hier beschrijven we het protocol voor de menselijke fibroblast cellen te herprogrammeren met de vier herprogrammering factoren gekloneerd in GFP-bevattende retrovirale backbone 4. Met behulp van de volgende protocol, genereren we de menselijke iPSCs in 3-4 weken in de menselijke cultuur ESC staat. Human IPSC kolonies lijken op hESC &#39;in morfologie en aangeven dat de GFP als gevolg van retrovirale transgene silencing. IPSC kolonies mechanisch geïsoleerd onder een fluorescentie microscoie zich op een vergelijkbare manier als hESC &#39;s. In deze cellen zien wij de expressie van meerdere genen en pluripotentie oppervlaktemerkers.
