Schistosoma parasieten zijn bloed staartvinnen die naar schatting 200 miljoen mensen wereldwijd een infecteren. In chronische infectie met Schistosoma de ernstige pathologie, zoals leverfibrose en Splenomegalie, wordt veroorzaakt door de immuunreactie tegen het parasieteneitjes dan de parasiet zelf 2. Parasieteneitjes induceren Th2 respons gekenmerkt door de productie van IL-4, IL-5 en IL-13, de andere activering van macrofagen en de rekrutering van eosinofielen. We beschrijven hier de injectie van Schistosoma mansoni eieren als een model om parasiet-specifieke Th2 cytokine reacties in de longen en de drainerende lymfeklieren, de vorming van pulmonaire granulomen rondom het ei, en luchtwegontsteking te onderzoeken. Na intraperitoneale sensibilisatie en intraveneuze uitdaging, S. mansoni eieren worden vervoerd naar de longen via de longslagaders waar ze worden gevangen in het longparenchymdoor granulomen samengesteld uit lymfocyten, eosinofielen en alternatief geactiveerde macrofagen 3-6. In verband met granuloomvorming, ontsteking in de broncho-alveolaire ruimtes, uitbreiding van de drainerende lymfeklieren en CD4 T-cel activatie kan worden waargenomen. Hier hebben we details van het protocol voor het isoleren van Schistosoma mansoni eieren van besmette levers (gemodificeerd uit 7), sensibiliserende en uitdagend muizen, en het herstellen van de organen (broncho-alveolaire lavage (BAL), long-en drainerende lymfeklieren) voor analyse. We hebben ook vertegenwoordiger van histologische en immunologische gegevens en suggesties voor aanvullende immunologische analyse. Over het algemeen deze methode zorgt voor een in vivo model om helminth geïnduceerde immunologische responsen te onderzoeken in de longen, dat in grote mate van toepassing zijn op de studie van de Th2-inflammatoire ziekten, waaronder worminfecties, fibrotische ziekten, allergische ontsteking en astma. Voordelen van dit model voor de studie van type 2 ontstekingen in de longen zijn de reproduceerbaarheid van een krachtige Th2 ontstekingsreactie in de long en drainerende lymfeklieren, het gemak van de beoordeling van de ontsteking door histologisch onderzoek van de granulomen rondom het ei, en het potentieel voor lange termijn opslag van de parasiet eieren.
