Lipopolysaccharide (LPS) is een belangrijk onderdeel van Gram-negatieve bacteriën buitenste membranen. Het is een drieledige molecuul dat uit lipide A, die ingebed is in de buitenste membraan, een kern oligosaccharide en het herhalen O-antigeen eenheden die buiten uitstrekken vanaf het oppervlak van de cel 1, 2. LPS is een immunodominante molecule die belangrijk is voor de virulentie en Pathogenese van vele soorten bacteriën, zoals Pseudomonas aeruginosa, Salmonella species, en Escherichia coli 3-5, en verschillen in LPS O-antigen samenstelling vormen de basis voor serotypering stammen. LPS is betrokken bij de gehechtheid aan gastheercellen aan het begin van de infectie en biedt bescherming tegen complement-gemedieerde doden; stammen die LPS missen kunnen worden verminderd voor de virulentie 6-8. Daarom is het belangrijk om LPS te visualiseren name van klinische isolaten. Het visualiseren van LPS banding patronen en erkenning door middel van specifieke eenntibodies kan nuttig zijn hulpmiddelen om stam lijnen te identificeren en om de verschillende mutanten te karakteriseren. In dit verslag beschrijven we een hete waterige fenol werkwijze voor de isolatie en zuivering van LPS van Gram-negatieve bacteriële cellen. Dit protocol kan de extractie van LPS afstand van nucleïnezuren en eiwitten die interfereren met visualisatie van LPS dat optreedt bij kortere minder intensieve winningsmethoden 9. LPS deze wijze bereid kunnen worden gescheiden natriumdodecylsulfaat (SDS)-polyacrylamide gelelektroforese (PAGE) en direct gekleurd met koolhydraten / glycoproteïne vlekken of standaard zilverkleuring methoden. Veel anti-sera aan LPS bevatten antilichamen die kruisreageren met buitenste membraan eiwitten of andere antigene doelen die kunnen hinderen reactiviteit waargenomen na West-immunoblot van SDS-PAGE gescheiden ruwe cellysaten. Protease behandeling van ruwe cellysaten alleen is niet altijd een effectieve manier van verwijderen van dezeachtergrond met behulp van deze of andere visualisatie methoden. Verder kan uitgebreid protease behandeling in een poging om deze achtergrond verwijderen tot slechte kwaliteit LPS dat niet goed is opgelost door elk van de bovengenoemde methoden. Om deze redenen zijn wij van mening dat de volgende protocol, aangepast van Westpahl en Jann 10, is ideaal voor LPS extractie.
