Liksom alla negativ-strängade RNA-virus, är i genomet hos influensavirus förpackas i form av virala ribonukleoproteinpartiklar komplex (vRNP), i vilka den enkelsträngade genomet är inkapslade av nukleoprotein (NP) och associerad med den trimera polymeras komplex bestående av PA, PB1, och PB2 subenheter. Emellertid, i motsats till de flesta RNA-virus, influensavirus utföra virala RNA-syntes i kärnan av infekterade celler. Intressant nog använder virala mRNA-syntes cellulära pre-mRNA som primers, och det har föreslagits att denna process äger rum på kromatin 1. Interaktioner mellan virala polymeraset och värd-RNA-polymeras II, såväl som mellan nonylfenol och nukleosomer värdceller har också karaktäriserats 1,2. Nyligen framtagandet av rekombinanta influensavirus som kodar en En-strep-markör fuserad genetiskt till C-terminalen av den PB2-subenheten av det virala polymeraset (rWSN-PB2-Strep 3) har blisv beskrivits. Dessa rekombinanta virus tillåta reningen av PB2-innehållande komplex, inklusive vRNPs, från infekterade celler. För att erhålla renat vRNPs är cellkulturer infekterades, och vRNPs är affinitetsrenades från lysat härrörande från dessa celler. Har dock lys förfaranden som används hittills varit baserad på en steg tvättmedel lys, som trots närvaron av en allmän nukleas, ofta extrahera kromatin-bundna materialet enbart ineffektivt. Våra preliminära arbete föreslog att en stor del av nukleära vRNPs inte extraherades under traditionella cellys, och kunde därför inte vara affinitetsrenade. För att öka denna extraktion effektivitet och för att separera kromatin-bundet från icke-kromatin-bundna kärnvapen vRNPs, anpassade vi en stegvis subcellulär utvinning protokoll till influensa virus-infekterade celler. Kortfattat separerar detta förfarande först kärnorna från cellen och därefter extraherar lösliga nukleära proteiner (här benämnd &quot;nucleoplasmic&quot; fraktion).Den återstående olösliga kärnmaterial digereras sedan med Benzonase, en ospecifik DNA / RNA-nukleas, följt av två steg saltextraktion: först med användning av 150 mM NaCl (benämnd &quot;CH150&quot;), sedan 500 mM NaCl (&quot;ch500&quot;) (fig 1 ). Dessa salt extraktionssteg valdes baserat på vår iakttagelse att 500 mM NaCl var tillräcklig för att solubilisera över 85% av kärn-vRNPs ändå medge bindning av märkta vRNPs till affinitetsmatrisen. Efter subcellulär fraktionering av infekterade celler, är det möjligt att affinitetsrena PB2-märkta vRNPs från varje individuell fraktion och analysera deras protein-och RNA-komponenter med Western blöt och primerförlängning, respektive. Nyligen utnyttjade vi denna metod för att upptäcka att vRNP export komplex form under slutet av poäng efter infektion på kromatinet fraktionen extraheras med 500 mM NaCl (ch500) 3.
