<p class="jove_content"> Isolamento de células a partir da fonte primária é um passo necessário em muitos protocolos mais complexos. Miltenyi oferece kits para isolar as células de vários organismos, incluindo seres humanos, primatas não-humanos, ratos e, como descrevemos aqui, camundongos. Separação magnética de células baseado em grânulos permite tanto a seleção positiva (ou depleção de células), bem como seleção negativa. Aqui, demonstramos seleção negativa de CD4 ve intocadas ou nd + células T helper. Usando este protocolo padrão que normalmente purificar as células que estão ≥ 96% CD4 pura + / CD3 +. Este protocolo é usado em conjunto com a dissecação de protocolo e 2-Photon Imagens de linfonodos periféricos em camundongos publicado na edição de 7 de Jove, para purificação de células T e outros tipos de células para adotivamente transferência para fins de imagem.
