<p class="jove_content"> Aislamiento de las células de la fuente primaria es un paso necesario en muchos protocolos más complejos. Miltenyi ofrece kits para aislar las células de varios organismos incluyendo los humanos, primates no humanos, ratas y, como se describe aquí ratones. Con bolas magnéticas basadas en la separación de células permite, ya sea para la selección positiva (o agotamiento de las células), así como la selección negativa. Este sentido, demuestran la selección negativa de CD4 he tocado ni na + células T colaboradoras. El uso de este protocolo estándar que suele purificar las células que son CD4 ≥ 96% puro + / CD3 +. Este protocolo se utiliza en conjunción con el protocolo de disección y 2-Photon imagen de los ganglios linfáticos periféricos en ratones publicado en el número 7 de Jove, para la purificación de las células T y otros tipos de células para la transferencia adoptiva con fines de imagen. A pesar de que no demostró el análisis FACS en este video de protocolo, se recomienda comprobar la pureza total de células aisladas con los anticuerpos apropiados a través de la FACS. Además, se demuestra que el método no estéril de aislamiento de células T. Si las células estériles son necesarios para su particular aplicación de usuario final, asegúrese de hacer todos los procedimientos se demuestra en la capilla de cultivo de tejidos en condiciones estándar de estéril.
