<p class="jove_content"> Isolering av celler från den primära källan är ett nödvändigt steg i många mer komplexa protokoll. Miltenyi offers kit att isolera celler från flera olika organismer, inklusive människor, icke-mänskliga primater, råtta och som vi beskriver här, möss. Magnetisk pärla-baserade cellseparation tillåter antingen positiv selektion (eller cellbristen) samt negativt urval. Här visar vi negativt urval av orörda eller Na har CD4 + hjälpar T-celler. Med denna standardprotokoll vi normalt renar celler som är ≥ 96% ren CD4 + / CD3 +. Det här protokollet används tillsammans med det protokoll Dissection och 2-Photon Imaging för perifera lymfkörtlar på möss publicerades i nummer 7 av Jove, för rening av T-celler och andra typer cell till adoptively överföring för avbildning ändamål. Även om vi inte visade FACS analys i detta protokoll videon, är det starkt rekommenderas att kontrollera den totala renheten i isolerade celler med hjälp av lämpliga antikroppar via FACS. Dessutom visar vi den icke-steril metod för T-cell isolering. Om sterila celler behövs för just din tillämpning hos slutanvändarna, se till att göra allt de visat förfaranden inom vävnadsodling huven under normala sterila förhållanden.
