Hjernesvulster er blant de mest aggressive og heterogene kreft kjent hos mennesker. Selv om deres tidligere oppdagelse og diagnose er tilrettelagt av moderne nevro-imaging teknologi, vi mangler fortsatt helbredende terapi for mange hjernesvulster, spesielt for diffuse, invasive eller de ligger dypt i hjernen. Hjernesvulster representerer den ledende årsaken til kreft dødelighet hos barn på grunn av deres svært aggressiv og ofte uhelbredelig natur. Glioblastoma (GBM), den vanligste primær hjernesvulst hos voksne, er en av de mest aggressive humane cancertyper fryktet for sine jevnt dødelig prognose 1. Denne svært ondartet astrocytic tumor (WHO grad 4) oppstår vanligvis i de cerebrale halvkuler av voksne, og kan også forekomme i små barn og spedbarn. Veksten er rask og infiltrerende og diagnostiske patologiske funksjoner inkluderer kjernekraft pleomorphism, mikrovaskulær spredning, og nekrose 2,3. Ved anvendelsen av stamceller biologi paradigmer til studiet av hjernesvulster, vi var de første forskerne som prospektivt identifisere og rense en undergruppe av celler fra menneskelige GBMS som utstilt stamcelleforskningen egenskaper spredning, selvfornyelse og differensiering in vitro 4 og i vivo 5. Vi søkte dyrkningsbetingelser og analyser opprinnelig brukt for å karakterisere normale nevrale stamceller (NSCs) in vitro 6,7 til flere barn og voksne hjernesvulster, og beriket for disse stilk-lignende celler ved celle sortering for den nevrale stamceller overflate markør CD133 8 ,9. Den CD133 + hjernesvulst brøkdel inneholdt celler som hadde en mye høyere frekvens av startfasen enn CD133-fraksjonen i NOD-SCID mus hjerner 5,10. Hjernesvulster er vanligvis består av morfologisk forskjellige celler som uttrykker en rekke nevrale avstamning markører. Bare en forholdsvis liten brøkdel av celler i tumoren med stilk celleegenskaper betegnes hjernesvulst initierende celler (BTICs), ha en evne til å differensiere langs flere linjer, selv-fornyelse og initiere svulster in vivo. Vi søkte dyrkningsbetingelser opprinnelig brukt for normale nevrale stamceller (NSCs) til en rekke menneskelige hjernesvulster og funnet ut at denne kulturen metoden spesielt velger for stilk-lignende populasjoner. Serumfritt medium (NSC) muliggjør opprettholdelse av en udifferensiert stamcelle tilstand, og tillegg av bFGF og EGF tillater spredning av multi-potente, selv-fornyelse, og utvidbare tumorspheres. For ytterligere å karakterisere hver svulsten BTIC befolkningen, vurderer vi celleoverflaten markører ved flowcytometri. Vi kan også sortere bestander av interesse for mer spesifikke characterizasjon. Selvfornyelse analysene er utført på enkelt BTICs sortert i 96 brønners plater; dannelsen av tumorspheres etter inkubering ved 37 ° C indikerer nærværet av en stilk eller stamceller. Flere celle tall av en bestemt populasjon kan også sorteres i forskjellige brønner for begrensende fortynning analyse, å analysere selvfornyelse kapasitet.
