Os tumores cerebrais estão entre os cânceres mais agressivos e heterogênea conhecidos em humanos. Embora a sua detecção precoce e diagnóstico ter sido facilitada pela tecnologia neuro-imagem moderna, ainda não temos terapias curativas para muitos tumores cerebrais, particularmente para difusas, as invasoras ou os localizados no fundo do cérebro. Os tumores cerebrais representam a principal causa de mortalidade por câncer em crianças devido à sua natureza altamente agressiva e muitas vezes incurável. Glioblastoma (GBM), o tumor cerebral primário mais comum em adultos, é um dos cancros mais agressivos humanos, temidos por seu prognóstico uniformemente fatal 1. Os tumores cerebrais são tipicamente constituídos por células morfologicamente diferentes que expressam uma variedade de marcadores de linhagem neural. Apenas uma fracção relativamente pequena de células no tumor com propriedades de células estaminais, células cerebrais chamados de tumores de iniciação (BTICs), possuir uma capacidade de se diferenciar ao longo de várias linhagens, de auto-renovação e iniciar tumores in vivo. Aplicou-se as condições de cultura normais, inicialmente utilizadas para as células estaminais neurais (NSC) para uma variedade de tumores cerebrais humanos e verificou que este método de cultura, especificamente selecciona para a haste-como populações. Meio isento de soro (NSC) permite a manutenção de um estado indiferenciado de células estaminais, e a adição de bFGF e EGF permite a proliferação de multi-potentes, de auto-renovação e tumorspheres expansíveis. Para caracterizar adicionalmente população BTIC cada tumor, avaliamos marcadores de superfície celular por citometria de fluxo. Podemos também classificar as populações de interesse para characteriz mais específicosção. A auto-renovação ensaios são realizados em BTICs individuais ordenadas em placas de 96 poços, a formação de tumorspheres após a incubação a 37 ° C, indica a presença de uma haste ou de células progenitoras. O número de células múltiplas de uma dada população podem ser classificados em diferentes poços para análise de diluição limitante, para analisar a capacidade de auto-renovação. Podemos também estudar a expressão diferencial de genes dentro de uma determinada população celular, utilizando células único de RT-PCR. Os protocolos que se seguem descrevem os processos para a dissociação e cultura de amostras humanas primárias para enriquecer as populações BTIC, bem como a dissociao de tumorspheres. Também incluídos estão os protocolos para coloração por análise de citometria de fluxo ou triagem, a auto-renovação ensaios e única célula de RT-PCR.
