Los tumores cerebrales son algunos de los cánceres más agresivos y heterogéneos conocidos en los seres humanos. A pesar de su detección temprana y el diagnóstico han sido facilitados por la moderna tecnología de neuroimagen, todavía carecemos de terapias curativas para muchos tumores cerebrales, en particular para los difusas, invasivos o localizados profundamente en el cerebro. Los tumores cerebrales representan la principal causa de mortalidad por cáncer en los niños debido a su naturaleza altamente agresivo e incurable a menudo. El glioblastoma (GBM), el tumor cerebral primario más común en los adultos, es uno de los cánceres humanos más agresivos, temían por su pronóstico uniformemente fatal 1. Los tumores cerebrales son típicamente compuesto de células morfológicamente distintas que expresan una variedad de marcadores de linaje neural. Sólo una fracción relativamente pequeña de las células en el tumor con propiedades de células madre, denominadas células tumorales cerebrales iniciador (BTICs), poseen una capacidad de diferenciarse a lo largo de múltiples linajes, auto-renovación, e iniciar los tumores in vivo. Se aplicaron condiciones de cultivo usadas originalmente para normales células madre neurales (NSC) a una variedad de tumores cerebrales humanos y se ha encontrado que este método de cultivo selecciona específicamente para como vástago-poblaciones. Medio libre de suero (NSC) permite el mantenimiento de un estado indiferenciado de las células madre, y la adición de bFGF y EGF permite la proliferación de multi-potente, auto-renovación, y tumorspheres expandibles. Para caracterizar aún más la población BTIC cada tumor, se evalúa marcadores de superficie celular por citometría de flujo.
