Men när man startar med tidig embryon, cellantalen tillräckliga för analyser är svårt att få med FACS, vilket komplicerar isolering av tidiga NC populations från enskilda embryon. Här beskriver vi ett förhållningssätt som inte är beroende av FACS och resulterar i en cirka 96% ren NC befolkningen baserad på en Wnt1-Cre aktiverad härkomst reporter 23. Metoden som presenteras här är anpassad från protokoll optimerade för odling av rått NC 11,13. Fördelarna med detta protokoll jämfört med tidigare metoder är att 1) ​​cellerna inte odlas på en feeder lager, 2) inte krävs för att få en relativt ren NC befolkning, 3) premigratory NC cellerna isoleras och 4 FACS) resultat är lätt kvantifieras.
