Under utviklingen neural crest (NC)-deriverte nevrale stamceller vandrer bort fra nevralrøret å danne autonome ganglier i viscerale organer som tarm og nedre urinveier. Både under utvikling og i modne vev disse cellene er ofte spredt over hele vev slik at isolering av separate populasjoner med metoder som laser capture mikro-disseksjon er vanskelig. De kan imidlertid være direkte visualisert ved uttrykk av fluorescerende reportere fordrevet fra regulatoriske regioner av nervecellen-spesifikke gener som Tyrosin hydroksylase (TH). Vi beskriver en metode optimalisert for høye avlinger av levedyktige TH + nevrale stamceller fra foster mus viscerale vev, inkludert tarmen og nedre urogenitaltractus (LUT), basert på dissosiasjon og fluorescens-aktivert celle sortering (FACS). Den Th genet koder hastighetsbegrensende enzym for produksjon av katekolaminer. Enteriske nevrale stamceller begynne å uttrykke TH durbehørig migrasjon i fosterstilling tarmen 1 og TH er også tilstede i en undergruppe av voksne bekken ganglia nerveceller 2-4. Vi avbildes uttrykk for dette transgenet i fosterets mus å dokumentere fordelingen av TH + celler i å utvikle LUT på 15,5 dager etter samleie (DPC), utpeking morgenen av plug påvisning som 0,5 DPC, og observert at en undergruppe av nevrale stamceller i fortetting bekken ganglier ekspress EGFP. For å isolere LUT TH + nevrale stamceller, optimalisert vi metoder som opprinnelig ble brukt til å rense neural crest stamceller fra foster mus tarmen 2-6. Tidligere forsøk på å isolere NC-deriverte populasjoner meget pålitelig ifordøyelse med en cocktail av collagenase og trypsin å få cellesuspensjoner for flowcytometri. I våre hender disse metodene produsert cellesuspensjoner fra LUT med relativt lav levedyktighet. Gitt den allerede lave forekomsten av nevrale stamceller i fosterlivet LUT vev, setter vi ut for å optimalisere dissosiasjon metoder slik at celle overlevelse i de endelige dissosierer skulle økes. Vi fastslått at milde dissosiasjon i Accumax (Innovative Cell Technologies, Inc), manuell filtrering, og flyt kildesortering lavtrykkene tillot oss å oppnå gjennomgående større overlevelse (&gt; 70% av totalt antall celler) med påfølgende avkastning av nevrale stamceller tilstrekkelige for nedstrøms analyse.
