Hjernen ventrikulære systemet er konservert blant virveldyr og er sammensatt av en serie av sammenkoblede hulrom kalt hjernen ventriklene, som danner under de tidligste stadiene av utvikling av hjernen og vedlikeholdes gjennom dyrets liv. For å undersøke regulering av permeabilitet under sebrafisk hjernen ventrikkel inflasjon, har vi utviklet en ventrikulær fargestoff oppbevaring analysen. Denne metoden bruker hjernen ventrikkel injeksjon i en levende sebrafisk embryo, en teknikk tidligere utviklet i vår lab 10, til fluorescently merke cerebrospinalvæsken. Embryoer blir deretter avbildet over tid som fluorescerende fargestoff beveger seg gjennom hjernen ventriklene og neuroepithelium. Avstanden fargestoff fronten beveger seg bort fra den basale (non-luminal) siden av neuroepithelium over tid er kvantifisert og er et mål på neuroepithelial permeabilitet (Figur 1). Vi observerer at fargestoffer 70 kDa og mindre vil bevege seg gjennom neuroepithelium og kan være detected utenfor embryonale sebrafisk hjernen ved 24 HPF (Figur 2).
