A. Wij beschrijven een werkwijze voor ex vivo kweken van Drosophila geheel hersenen. Dit kan gebruikt worden als een tegenhanger van chronische genetische manipulaties voor het onderzoeken van de celbiologie en de ontwikkeling van het centrum van hersenstructuren door toe te staan ​​acute farmacologische interventies en live beeldvorming van cellulaire processen. Als voorbeeld van de techniek vóór werken ons laboratorium 1 is gebleken dat een eerder opgenomen subcellulair compartiment ligt tussen het axonale en somatodendritisch compartimenten axonen van Drosophila centrale hersenen. De ontwikkeling van dit compartiment, aangeduid als axon eerste segment (AIS) 2 is getoond genetisch afhankelijk van de neuron-specifieke cycline afhankelijke kinase, Cdk5. We laten hier zien dat ex vivo behandeling van wild-type Drosophila larven hersenen met de Cdk5-specifieke farmacologische remmers roscovitine en olomoucine 3 acute veranderingen in het actine organisatie veroorzaakt, enin de lokalisatie van de cel-oppervlakte-eiwit Fasciclin 2, dat de veranderingen gezien in mutanten die Cdk5 activiteit missen genetisch na te bootsen. Een tweede voorbeeld van de ex vivo cultuur techniek is bedoeld voor herinrichting van de aansluitingen van de embryonale paddestoel lichaam (MB) gamma-neuronen tijdens metamorfose van larve tot volwassen. De paddestoel lichaam is het centrum van olfactorische leren en geheugen in de vlieg 4, en deze gamma-neuronen hun axonale en dendritische takken te snoeien tijdens de pop ontwikkeling en vervolgens weer uit te breiden takken op een later tijdstip bepaald aan de volwassen innervatie patroon 5 vast te stellen. Snoeien van deze neuronen van de MB is aangetoond gebeuren door plaatselijke degeneratie van neurite takken 6, door een mechanisme dat wordt geactiveerd door ecdyson een steroid hormoon, die op de ecdyson receptor B1 7 en is afhankelijk van de activiteit van de ubiquitine-proteasoom-systeem 6. Onze methode van ex vivo kweken kan be gebruikt om verder te ondervragen het mechanisme van ontwikkeling remodeling. We hebben gevonden dat in de ex vivo cultuur instelling gamma neuronen van de MB het proces van ontwikkeling snoeien samengevat een tijdsverloop vergelijkbaar met die in vivo. Het was echter van essentieel belang om tot 1,5 uur na puparium vorming wachten voordat explanteren van het weefsel, zodat de cellen onherstelbaar te zetten voor metamorfose, dissectie van dieren bij het begin van de verpopping leidde tot weinig of geen metamorfose in cultuur. Zo, met de juiste modificatie, kan de ex vivo cultuur aanpak worden toegepast om te studeren dynamische en steady-state aspecten van de centrale hersenen biologie.
