Onze protocol voor de isolatie van muizen AECII door flowcytometrie biedt een snelle manier toegang primaire cellen uit de muizenlong voor een hele reeks van functionele en moleculaire studies. In de afgelopen jaren, is de bijdrage van alveolaire type II epitheelcellen (AECII) diverse aspecten van immuunregulatie in de longen vaker erkend. AECII is aangetoond dat deel cytokine productie in ontstoken luchtwegen en zelfs als antigen-presenterende cellen in zowel infectie en T-cel gemedieerde auto-immuniteit 1-8. Daarom zijn ze bijzonder interessant ook binnen klinische vraagstellingen, zoals hyperreactiviteit van de luchtwegen bij buitenlandse en zelf-antigenen alsook infecties die direct of indirect richten AECII. Echter ons begrip van de gedetailleerde immunologische functies bediend door alveolaire type II epitheelcellen van gezonde long en in ontsteking blijft fragmentarisch. Veel studies over AECII functie worden uitgevoerd met behulp van de muis of mens alveolaire epitheliale cellijnen 9-12. Werken met cellijnen biedt zeker een aantal voordelen, zoals de beschikbaarheid van een groot aantal of cellen voor uitgebreide analyses. Echter, wij geloven het gebruik van primaire muriene AECII zorgt voor een beter begrip van de rol van dit type cel in complexe processen, zoals een infectie of auto-immuun ontsteking. Primaire murine AECII direct geïsoleerd uit dieren die lijden aan dergelijke ademhalingsproblemen, wat betekent dat ze zijn onderworpen aan de aanvullende extrinsieke factoren een rol spelen in de geanalyseerde setting. Als voorbeeld kan levensvatbare AECII worden geïsoleerd uit muizen intranasaal geïnfecteerd met influenza A-virus, die voornamelijk deze cellen richt voor replicatie 13. Belangrijker door ex vivo infectie van AECII geïsoleerd uit gezonde muizen, kan studies van de cellulaire responsen bevestigd na infectie worden verlengd. Onze protocol voor de isolatie van primaire murine AECII is gebaseerd op enzymatische afbraak van de muizenlong gevolgd door etikettering van de resulterende celsuspensie met antilichamen specifiek voor CD11c, CD11b, F4/80,CD19, CD45 en CD16/CD32. Granulaire AECII worden vervolgens geïdentificeerd als ongelabelde en zijwaartse verstrooiing hoog (SSC hoog) celpopulatie worden gescheiden door fluorescentie geactiveerde celsortering 3. In vergelijking met andere werkwijzen voor het isoleren primaire epitheelcellen muizenlongen, ons protocol voor flowcytometrische isolatie van AECII door negatieve selectie levert ongerepte zeer levensvatbare en zuivere AECII in relatief korte tijd. Bovendien, in tegenstelling tot conventionele methoden van isolatie door panning en uitputting van lymfocyten via binding van antilichaam-gekoppelde magnetische korreltjes 14, 15, flowcytometrische cell-sortering maakt onderscheid door celgrootte en granulariteit. Aangezien instrumentatie voor flowcytometrische celsortering beschikbaar is, kan de beschreven procedure worden toegepast tegen relatief lage kosten. Naast standaard antilichamen en enzymen voor long desintegratie geen extra reagentia zoals magnetischebeads vereist. De geïsoleerde cellen zijn geschikt voor diverse functionele en moleculaire studies, waaronder in vitro kweek en T-cel stimulatie assays en transcriptoom, proteoom of secretoom analyses 3, 4.

Bij het sorteren longcelsuspensies geïsoleerd uit gezonde muizen, zal de AECII poort doorgaans goed zijn voor ongeveer 42 ± 10% van alle gebeurtenissen. Dit percentage kan aanmerkelijk lager bij muizen met ademhalingsaandoeningen zoals een virale infectie worden gebruikt als de oorspronkelijke celsuspensie een aanzienlijk hoger percentage lymfocyten en andere cellen aangetrokken om de luchtwegen bevatten.
