Optogenetics is een krachtige nieuwe techniek die ongekende controle over specifieke neuronale subtypes mogelijk maakt. Dit kan worden benut moduleren neurale circuits met anatomische en temporele precisie, maar worden de celtype willekeurige en invasieve effecten van elektrische stimulering door een elektrode. Implantatie van glasvezel zorgt voor consistente, chronische stimulering van neurale circuits meerdere sessies in wakker gedragen muizen met minimale schade aan weefsel. Het ophelderen van de patronen van neuronale connectiviteit is een uitdaging voor zowel de klinische en fundamentele neurowetenschappen. Elektrofysiologie is de gouden standaard voor het analyseren van patronen van synaptische connectiviteit, maar gekoppeld elektrofysiologische opnames kunnen zowel omslachtig en experimenteel te beperken. De ontwikkeling van optogenetics introduceert een elegante methode om neuronen en circuits stimuleren, zowel in vitro en in vivo een 2,3. Door gebruik celtype specifieke promoter activiteit opsin expressie in discrete neuronale populaties kan men nauwkeurig stimuleren genetisch gedefinieerde neuronale subtypes in verschillende circuits 4-6. Goed beschreven methoden neuronen, inclusief elektrische stimulatie en / of farmacologische manipulaties, stimuleren vaak willekeurig celtype, invasieve en kunnen beschadigen omliggende weefsels. Deze beperkingen kunnen veranderen normale synaptische functie en / of circuit gedrag. Bovendien wegensde aard van de manipulatie, de huidige methoden vaak acute en terminal. Optogenetics biedt de mogelijkheid om neuronen te stimuleren in een relatief onschadelijke wijze en in genetisch gerichte neuronen. De meeste studies met in vivo optogenetics momenteel een optische vezel geleid via een canule geïmplanteerd 6,7, maar beperkingen van deze methode zijn beschadigd hersenweefsel met herhaalde inbrengen van een optische vezel en potentiële breuk van de vezel binnen de canule. Gezien de snel groeiende gebied van de optogenetics, een meer betrouwbare methode van chronische stimulatie is noodzakelijk om op lange termijn studies te vergemakkelijken met een minimale collaterale weefselbeschadiging. Hier geven we onze gemodificeerd protocol als video artikel de werkwijze effectief en elegant beschreven in Sparta et al. vullen. 8 voor de vervaardiging van een glasvezel implantaat en zijn vaste bevestiging op de schedel van verdoofde muizen, en de montage van de fiberoptische koppeling verbinden van het implantaat aan een lichtbron. Het implantaat, verbonden met optische vezels om een solid-state laser, maakt een efficiënte methode om functionele neuronale circuits chronisch photostimulate met minder weefselschade 9 met kleine, afneembare tethers. Permanente bevestiging van de glasvezel implantaten zorgt voor een consistente, op lange termijn in vivo optogenetic studies van neuronale circuits in wakker, gedragen muizen 10 met minimale weefselbeschadiging.
