1. L&#39;immunothérapie du cancer permet d&#39;exploiter la spécificité de la réponse immunitaire à cibler et à éliminer les tumeurs. Thérapie cellulaire adoptive (ACT) basé sur le transfert adoptif de lymphocytes T génétiquement modifiées pour exprimer un récepteur d&#39;antigène chimérique (CAR) a montré très prometteur dans les essais cliniques 1-4. Il ya plusieurs avantages à utiliser CAR + lymphocytes T pour le traitement de cancers, y compris la capacité de cibler des antigènes du CMH non réglementées et pour fonctionnaliser les lymphocytes T pour la survie optimale, tête chercheuse et de la persistance dans l&#39;hôte, et enfin à induire l&#39;apoptose de CAR + Les cellules T dans le cas de 5 toxicité hôte. La délimitation des fonctions optimales de CAR + lymphocytes T associés à un bénéfice clinique est essentielle pour concevoir la prochaine génération des essais cliniques. Les progrès récents de l&#39;imagerie des animaux vivants comme la microscopie multiphotonique ont révolutionné l&#39;étude de la fonction des cellules immunitaires in vivo 6,7. Bien que ces études ont fait progresser notre compréhension des fonctions des cellules T in vivo, des cellules T ACT basée sur des essais cliniques nécessite la nécessité de relier les caractéristiques moléculaires et fonctionnelles des cellules T préparations pré-infusion avec une efficacité clinique après la perfusion, en utilisant en tests in vitro des cellules T surveillance des fonctions telles que, la cytotoxicité et la sécrétion de cytokines. En combinaison avec la microscopie à fluorescence automatisé time-lapse, quoiqueusands d&#39;effecteur-cible interactions peuvent être surveillés simultanément par des puits individuels d&#39;imagerie d&#39;un tableau nanowell. Nous présentons ici une méthode à haut débit pour la surveillance cytotoxicité à médiation cellulaire T-au niveau d&#39;une seule cellule qui peut être largement appliquée à l&#39;étude de la fonctionnalité des lymphocytes T cytolytiques.

Un exemple de l&#39;application de l&#39;analyse à haut débit cytolytique est démontré dans la figure 2. En bref, étiqueté CD19 spécifique CAR + cellules T ont été co-incubées avec des cellules de souris marqués cibles EL4 dans les puits individuels d&#39;un tableau nanowell (articles 1-5).
