Over de anesthesie: Aangezien in utero elektroporatie in de hypothalamus technisch lastig en vereisen een langere tijd narcose we bij voorkeur inductie en handhaven van verdoving door toediening van een mengsel van zuurstof en isofluraan. In onze ervaring, kunnen dieren goed verdoofd blijven op deze manier voor een periode van maximaal een uur op zijn minst, het herstel van de moeder is erg snel, en de overleving van embryo&#39;s verbeterd. Genetische manipulatie van de embryonale hersenen van muizen is een geprefereerde aanpak te leren over ontwikkelingsregulatie. De generatie van mutante muizenlijnen echter traag en duur. Een methode wel specifieke genetische veranderingen in ontwikkeling neuronen van de hersenen van zoogdieren introduceren in utero elektroporatie. In wezen de techniek bestaat het transfecteren DNA in de embryonale hersenen neuroepithelium door middel van elektrische pulsen, waarna men het embryo te overleven gedurende een bepaalde tijd, het verzamelen van de hersenen en onderzoek ze mogelijke nieuwe, informatief fenotypes. Hierdoor kan de experimentator vrijwel direct testen hypotheses zonder lange wachttijden voor het samenstellen van muismutanten. Transfectie van DNA in ontwikkelende embryo&#39;s begonnen met in-ovo elektroporatie op kippenembryo&#39;s 1. De essentiële proof-of-concept voor de muis werd uitgevoerd in de cultuur <sup&gt; 2. Dit werd al snel gevolgd door de eerste beschrijvingen van de techniek op de muis in de baarmoeder 3,4. Het belangrijkste probleem is de hersenen van embryo ontwikkeling in utero transfecteren zonder hen of moeder doden. Leren om de noodzakelijke operatie uit te voeren (laparotomie, injectie, elektroporatie) vereist een lange trainingsperiode. Zodra de operatie is de knie tot het punt waar de embryo overleving verhouding aanvaardbaar is, de volgende belangrijke vraag is: welke hersenstructuren toegankelijk zijn? Niet verrassend, de eerste gepubliceerde artikelen tonen de resultaten verkregen met in utero elektroporatie gericht op de corticale ontwikkeling 5-9. Dit is nog steeds geldt voor de meeste publicaties met deze techniek, omdat het gebied van de ontwikkeling van muizenhersenen meest toegankelijke chirurgische procedures is de cortex (Figuur 1). Deze regio blijft een moeilijke uitdaging voor onderzoekers verbonden aan genetisch manipuleren van de hersenen van muizen in de baarmoeder. Voor zover wij weten, melden slechts zeer weinig artikelen over de resultaten van de in utero transfectie in de muis hypothalamus 20,21. De functionele betekenis van de hypothalamus cannot worden overdreven, omdat het reguleert gedrag zoals eten en drinken, paring, fokken en opvoeden 22. Bovendien, veranderingen in hypothalamus ontwikkeling helpen de latere leeftijd aandoeningen zoals zwaarlijvigheid, hypertensie, diabetes en voortijdige puberteit 23 ontstaan. In staat zijn om genetisch veranderen de hypothalamus tijdens de ontwikkeling zou zorgen voor een zeer krachtig instrument om het te begrijpen. De basis chirurgische protocol voor laparotomie zwangere muizen die we hier gebruikt is vergelijkbaar met die in andere protocollen 11,13,14,24. We zullen ze hier kort beschrijven op volledigheid. Sleutel tot onze procedure, anderzijds, zijn de vorm van anesthesie, de plaats van toediening, de aard van de elektroden en de insertie en de positie van de positieve elektrode ten opzichte van het hoofd van de embryo&#39;s. We verkiezen voor inductie en handhaven verdoving door inhalatie gas via eenvoudige intraperitoneale anesthesie, aangezien de eerste maakt deiets langere narcose vereist een moeilijke operatie. Isofluraan inhalatie resulteert in sneller herstel van anesthesie, aangezien meestal de moeder toont normaal gedrag heeft minuten na de operatie. De eenvoudigste injectiepunt van de DNA-oplossing met de glazen micropipet is de laterale ventrikel, die echter volledig ongeschikt voor hypothalamus elektroporatie. Injectie rechtstreeks in het derde ventrikel is inderdaad cruciaal voor diepe diencephalic structuren te richten. Het is mogelijk om de hypothalamus transfecteren van E12.0 en E12.5 met standaard, off-the-shelf elektroden. We hebben een aantal van de elektroden vervaardigd door Nepa Gene (Chiba, Japan) bijzonder geschikt voor dit doel gevonden. Met onze procedure transfectie van de gehele hypothalamus neuro-epitheel of gedeeltelijke, regionale transfectie afhankelijk elektrode oriëntatie krijgen we. Hier laten we de techniek door transfecteren van de mammillary lichaam, misschien welde diepste en meest verzonken van alle hypothalame kernen. Daarnaast tonen we gedetailleerde histologische analyse van de getransfecteerde cellen naar het cellulaire niveau van de resolutie. Een vergelijking van de in utero elektroporatie met andere benaderingen transfecteren van de muis ontwikkelende hersenen in utero te vinden in de sectie discussie.

Genetische modificatie van specifieke gebieden van de ontwikkelende hersenen van zoogdieren is een zeer krachtige experimentele benadering. Echter, het genereren van nieuwe muismutanten is vaak frustrerend traag. Er is aangetoond dat de toegang tot de hersenen van muizen ontwikkelen in utero met redelijke post-operatory overleving mogelijk. Toch resultaten met deze procedure zijn bijna uitsluitend gerapporteerd voor de meest oppervlakkige en gemakkelijk toegankelijk deel van de ontwikkeling van de hersenen, dat wil zeggen de cortex. De thalamus, een smaller en middengebied is moeilijker doel gebleken. Transfectie in diepere kernen, met name die van de hypothalamus, is misschien wel de meest uitdagende en dus zeer weinig resultaten zijn gemeld. Hier laten we een procedure om de gehele hypothalamus neuroepithelium of deel daarvan (hypothalamische regio) voor transfectie door elektroporatie richten. De sleutels van onze aanpak zijn langer narcose keer, injectie in de third ventrikel en passende vorm en plaatsing van de elektroden. Daarnaast tonen we de resultaten van targeting en daaropvolgende histologische analyse van de meest verzonken hypothalamus nucleus, het mammillary lichaam.

De meeste hypothalamus neuronen zijn geboren tussen E11.5 tot E15.2, volgens geboorte-dating-analyse bij de rat 26 vertaald in de wat kortere muis ontwikkeling 27,28. De piek van de hypothalamus neurogenese wordt bereikt bij E12.5 29-31. Dienovereenkomstig, op de leeftijd transfectie gekozen voor deze studie (E12.5), een groot deel van neuronen van de hypothalamus codeerbaar op elk rostro-caudale niveau.
