Genetisk manipulation av embryonala mus hjärnan är en föredragen metod för att lära sig om utvecklings-reglering. Alstring av mutanta mus linjer är emellertid långsam och dyr. En kraftfull metod för att introducera specifika genetiska förändringar i utvecklingen av nervceller i däggdjur hjärnan är i livmodern elektroporering. Emellertid den funktionella betydelsen av hypothalamus cannot överskattas, eftersom det reglerar beteenden som att äta och dricka, parning, avel och föräldraskap 22. Dessutom förändringar i hypotalamus utveckling bidrar till att härröra senare i livet tillstånd som fetma, högt blodtryck, diabetes och för tidig pubertet 23. Att kunna ändra genetiskt hypotalamus under utveckling skulle ge ett mycket kraftfullt verktyg för att förstå det. Den grundläggande kirurgiska protokoll för laparotomi av dräktiga möss som vi använder här är liknande den som används i andra protokoll 11,13,14,24. Vi kommer att beskriva dem här kort för fullständighet. Nyckeln till vårt förfarande, å andra sidan, är den typ av anestesi, platsen för injektionen, typen av elektroder och införande och placering av den positiva elektroden i förhållande till embryots huvud. Vi föredrar att inducera och underhålla anestesi genom gas inandning över enkel intraperitoneal anestesi, eftersom den förstnämnda möjliggörnågot längre narkos krävs för en svår operation. Isofluran inandning resulterar i snabbare återhämtning från anestesi, eftersom vanligtvis modern demonstrerar normalt beteende redan minuter efter operationen. Det enklaste punkten för injektion av DNA-lösningen med glasmikropipett är den laterala ventrikeln, som dock är helt olämpliga för hypotalamus elektroporering. Injektion direkt i den tredje ventrikeln är verkligen avgörande att rikta djupa diencephalic strukturer. Det är möjligt att transfektera hypotalamus från E12.0 eller E12.5 med standard, off-the-shelf elektroder. Vi har funnit en del av elektroderna som tillverkas av Nepa Gene (Chiba, Japan) särskilt lämpad för detta ändamål. Med vårt förfarande erhåller vi transfektion av hela hypotalamus neuroepitelet eller delvis, regional transfektion beroende på elektrod orientering. Här kan vi demonstrera tekniken genom transfektion av mammillary kroppen, utan tvekanden djupaste och mest infälld allt hypotalamus kärnor. Dessutom visar vi detaljerad histologisk analys av de transfekterade cellerna ner till cellnivå av upplösning. En jämförelse av i livmodern elektroporering med andra metoder för transfektion musen utvecklar hjärnan i livmodern kan hittas i diskussionen avsnittet.

Genetisk modifiering av specifika regioner i tredje däggdjurshjärnan är en mycket kraftfull experimentell metod. Men generera mutanter nya mus är ofta frustrerande långsamt. Det har visats att tillgången till mushjärnan utvecklas i livmodern med rimlig post-operationslampan överlevnad är möjlig. Ändå har resultat med detta förfarande har rapporterats nästan uteslutande i de mest ytliga och lättillgänglig del av den växande hjärnan, dvs cortex. Thalamus, ett smalare och mer medial region, har visat sig vara svårare att rikta. Transfektion in djupare kärnor, speciellt de av hypotalamus, är kanske den mest utmanande och därför mycket få resultat har rapporterats. Här visar vi ett förfarande för att rikta hela hypotalamus neuroepitelet eller en del av det (hypotalamus regioner) för transfektion genom elektroporation. Nycklarna till vårt tillvägagångssätt är längre narkos gånger, injektion i tREDJE ventrikeln, och lämplig typ och positionering av elektroderna. Dessutom visar vi resultaten av inriktning och efterföljande histologisk analys av de försänkta hypotalamus kärnan, mammillary kroppen.

De flesta hypotalamus nervceller föds mellan E11.5 till E15.2, enligt födelse-dejting analys hos råtta 26 översättas till något kortare musen utveckling 27,28. Toppen av hypotalamus neurogenesisen nås vid E12.5 29-31. Följaktligen, vid transfektion ålder valdes för denna studie (E12.5), kan en stor del av hypotalamus nervceller märkas vid varje given rostro-caudal nivå.
