the flybar: administering alcohol to flies

Moscas de frutas (Drosophila melanogaster) são um modelo estabelecido para a pesquisa de álcool e biologia circadiano. Recentemente, mostramos que o relógio circadiano modula sensibilidade ao álcool, mas não a formação de tolerância. Aqui, descrevemos nosso protocolo em detalhe. O álcool é administrado às moscas usando flybar. Nesta configuração, o vapor de álcool saturado é misturado com o ar umidificado em proporções definidas e administradas às moscas em quatro tubos simultaneamente. As moscas são criadas sob condições padronizadas, a fim de minimizar a variação entre as repetições. Três dias de idade moscas de diferentes genótipos ou tratamentos são utilizados para os experimentos, de preferência por moscas de dois pontos diferentes de tempo (por exemplo, CT 5 e CT 17) fazendo comparações diretas possível combinar. Durante a experiência, as moscas são expostos durante 1 hora a percentagem pré-determinada de vapor de álcool e o número de moscas que exibem a perda do reflexo de endireitamento (LORR) ou sedção são contados a cada 5 min. Os dados podem ser analisados ​​utilizando-se três abordagens estatísticas diferentes. A primeira é a de determinar o tempo em que 50% das moscas têm perdido o reflexo de endireitamento e usar uma análise da variância (ANOVA) para determinar se existem diferenças significativas entre pontos de tempo. O segundo é o de determinar as moscas percentuais que mostram Lorr após um determinado número de minutos, seguido de uma análise ANOVA. O último método é analisar toda a série vezes utilizando estatística multivariada. O protocolo também pode ser usado para as experiências não circadianos ou comparações entre genótipos.
