Utilizzando i protocolli descritti in questo documento è possibile estrarre e cultura spirale neuroni gangliari e di indagare laser evocata attività elettrica eseguendo interi esperimenti di patch clamp cellulari. Quando utilizzato in vitro, la tecnica patch clamp fornisce un livello di controllo sui parametri sperimentali che non è realizzabile in vivo. I campi della neurofisiologia e della bionica medica si basano molto sulle tecniche che consentono la stimolazione controllabile di risposte elettriche nei tessuti nervosi. Mentre la stimolazione elettrica rimane il gold standard nella eccitazione neurale, soffre di una serie di inconvenienti, quali la presenza di artefatti di stimolazione quando si registra risposte neurali, e una mancanza di stimolazione specificità dovuta alla diffusione di corrente nel tessuto circostante 1. Gli ultimi due decenni hanno visto lo sviluppo di tecniche di stimolazione otticamente mediate 2. Molte di queste tecniche richiede modificazione del tessuto bersaglio, tramite l&#39;aggiunta di una particolare molecola (molecole es gabbia) 3 o qualche forma di manipolazione genetica (es. optogenetics) 4, nessuno dei quali è facilmente applicabile al di fuori di un ambiente di ricerca. Di particolare interesse è dunque stimolazione neurale infrarossi (INS), whereby tessuto neurale è eccitato dalla luce laser infrarossa pulsata. INS ha il potenziale per superare molte delle carenze della stimolazione elettrica, consentendo altamente specifico, senza contatto stimolazione del tessuto neurale 2. Tuttavia, mentre INS è stata dimostrata con successo in una varietà di impostazioni in vivo, il meccanismo preciso di eccitazione rimane incerta. Alcune pubblicazioni recenti hanno dimostrato progressi verso scoprire il meccanismo che sta dietro INS 5-7. Rapido riscaldamento dovuto all&#39;assorbimento della luce laser da acqua sembra giocare un ruolo chiave. Tuttavia, al di là di questo un consenso deve ancora essere raggiunto. Shapiro et al. 7 propongono un meccanismo molto generale che riscaldamento rapido provoca una perturbazione nella distribuzione di particelle cariche adiacenti alla membrana cellulare, portando ad una variazione della capacità della membrana cellulare e conseguente depolarizzazione. In questa fase non è chiaro come questi meccanismi combinano, o anche se vi sono ulteriori fattori che sono ancora da identificare. Anche se un piccolo numero di pubblicazioni (riferimenti 5,7-9) hanno indagato INS in vitro, la stragrande maggioranza dei lavori pubblicati in questo campo è stata effettuata in vivo (ad es riferimenti 1,6,10-18). Stimolazione infrarossi dei neuroni uditivi è stata una zona di particolare interesse, a causa delle potenziali applicazioni in impianti cocleari 10,14-18. Mentre esperimenti in vivo sono importanti per verificare l&#39;efficacia della tecnica in varie impostazioni, l&#39;aumento del livello di controllo offerta da studi in vitro dovrebbe portare ad una comprensione più dettagliata del meccanismosmo responsabile INS. È stato dimostrato negli ultimi anni che pulsata, luce laser infrarosso può essere utilizzato per ottenere risposte elettrici nel tessuto neurale, indipendente da qualsiasi ulteriore modifica del tessuto bersaglio. Stimolazione neurale infrarossi è stato riportato in una varietà di tessuto neurale e sensoriali periferici in vivo, con particolare interesse mostrato nella stimolazione di neuroni nel nervo uditivo. Il protocollo presentato qui descrive un metodo di patch clamp cellula intera progettato per facilitare le indagini di stimolazione neurale infrarossi in colture di neuroni uditivi primari. Dal fondo caratterizzare la risposta di queste cellule di illuminazione laser infrarosso in vitro in condizioni controllate, può essere possibile ottenere una migliore comprensione del physica fondamentalel ed i processi biochimici sottostanti stimolazione neurale infrarossi.

Neuroni gangliari spirale rispondono al laser con illuminazione ripetibile in entrambe le forme d&#39;onda di tensione-clamp e configurazioni di registrazione corrente-clamp. Figura 3a mostra tipici cambiamenti nel flusso di corrente attraverso una membrana cellulare in risposta a un 2,5 msec, 0,8 mJ impulsi laser (risposta media da 6 impulsi laser, ripetuti ad intervalli di 1 sec) con il potenziale di membrana tenutosi a -70 mV, -60 mV e -50 mV.
