Enquanto a estimulação elétrica continua sendo o padrão ouro em excitação neural, sofre de uma série de inconvenientes, tais como a presença de artefatos de estímulo durante a gravação de respostas neurais, e uma falta de especificidade estimulação devido à propagação de corrente em um tecido circundante. Demonstrou-se em anos recentes que pulsado, o laser de infravermelhos podem ser utilizadas para induzir respostas eléctricas em tecido neural, independente de qualquer outra modificação do tecido alvo. Estimulação neural de infravermelho foi relatada numa variedade de tecido neural periférico e sensorial in vivo, com particular interesse mostrado na estimulação de neurónios do nervo auditivo. No entanto, enquanto que o INS foi mostrado para trabalhar nestas configurações, o mecanismo (ou os mecanismos) pelo qual a luz infravermelha provoca excitação neuronal não é actualmente bem compreendido. O protocolo aqui apresentado descreve um método de fixação de membranas de células inteiras concebido para facilitar a investigação de estimulação neural infravermelhos em cultura neurónios auditivos primários. Por caracterizar completamente a resposta destas células a iluminação laser infravermelho, in vitro, sob condições controladas, pode ser possível obter uma melhor compreensão da physica fundamentaisl e os processos bioquímicos subjacentes estimulação neural por infravermelhos.

Neurónios do gânglio espiral respondem à iluminação laser com formas de onda repetíveis em ambos tensão braçadeira e configurações de gravação actual-clamp. Figura 3a mostra alterações típicas na corrente de fluxo através de uma membrana celular, em resposta a um 2,5 ms, 0,8 mJ impulso laser (de resposta média de 6 pulsos de laser, repetido a intervalos de 1 segundo), com o potencial de membrana realizada a -70 mV, -60 mV e -50 mV.
