Regulatoire T-cellen (Tregs) zijn essentieel voor immunotolerantie aan zichzelf en aan bepaalde vreemde antigenen. Tregs kunnen vanuit naïeve CD4 T-cellen in vitro met TCR-en co-stimulering in de aanwezigheid van TGFp en IL-2. Dit draagt ​​enorm potentieel voor toekomstige therapie echter de moleculen en signaalwegen control differentiatie grotendeels onbekend. Primaire T-cellen kunnen worden gemanipuleerd door ectopische genexpressie, maar gemeenschappelijke methoden niet aan de belangrijkste naïeve toestand van de T-cel te richten voor primaire antigen herkenning.
