In diesem Protokoll beschreiben wir ein Verfahren zur Isolierung von pankreatischen Azinuszellen. Dieses Verfahren macht es möglich, mehr als 20 x 10 6 Zellen pro Tier acinar in weniger als 1 h zu isolieren. Dank seiner schnellen und einfachen Implementierung (so viele wie 10 Bauchspeicheldrüsen unabhängig pro Experiment parallel verarbeitet werden), wird dieses Protokoll als guter Kompromiss zwischen bestehenden Isolation Methoden 3-5,9-12,17. Ein häufig auftretendes Problem für Forschungslabore auf exokrinen Pankreasgewebe ist die Schwierigkeit der Kultivierung Azinuszellen in vitro für einen bestimmten Zeitraum lang genug, um ein langfristiges Experiment erlauben. Ein Faktor behindern Entwicklung solcher Systeme Kultur ist die intrinsische Empfindlichkeit von Pankreas-Gewebe experimentelle Manipulation durch den hohen Gehalt in glykolytischen, proteolytische und lipolytische Enzyme, die wörtlich verdauen Pankreasgewebe, wenn sie bei der Isolierung von pankreatischen Zellen freigesetzt werden. Ein zweiter Faktor ist die bemerkenswerte in vitro Plastizität Azinuszellen, die ihre sekretorischen Eigenschaften verlieren und transdifferenzieren anderen reifen Zellen, wie Pankreasgangzellen oder Leber-ähnlichen Zellen 1. Neigen In vitro variiert diese Zellplastizität mit den experimentellen Bedingungen (z. B. Mehrere Methoden zur Isolierung und Kultur der Azinuszellen, zuerst aus dem Meerschweinchen Bauchspeicheldrüse 3-5 entwickelt. Zunächst werden die Protokolle Verdauung von Pankreasgewebe mit Kollagenase, Chymotrypsin und einer Protease Cocktail beteiligt, mit ultimativen Isolierung durch kräftiges mechanische Dissoziation. Die Zellen der Bauchspeicheldrüse auf diese Weise isoliert angezeigt abnorme strukturelle und funktionelle Eigenschaften, insbesondere ein Verlust von apikalen Strukturen und erhebliche Schäden an ihrer Membran-Rezeptoren. Isolierte Zellen blieben nur für 1 oder 2 Tage lebensfähig. Herstellung von dispergierten acini unterhält ihre intra-und interzellulären Architektur, die Erhaltung Zellmembranen, Schadensbegrenzung zu Oberflächen-Rezeptoren und damit die Verbesserung der exokrinen Sekretion in Reaktion auf Sekretagoga 6-8. Als result, bietet dieses Verfahren den großen Vorteil, sich Azinuszelltumoren Lebensfähigkeit zu 7-10 Tagen in vitro. Darüber hinaus ist dieses Verfahren derzeit Azinuszelltumoren Isolierung 9-12 da die Wartung der interzellulären Kontakte, darunter Zelle Kopplung durch Gap Junctions, ist eine wesentliche Determinante des exokrinen Pankreas Azinuszelltumoren Phänotyp 13 bevorzugt. Da die Entdifferenzierung von Azinuszellen und ihre transdifferentiation zu duktalen Zellen ist einer der vorgeschlagenen Mechanismen für die Entstehung von aggressiven exokrinen Bauchspeicheldrüsenkrebs 14, ist die dispergierte acini Modell auch ein geeignetes System zur Pankreas-Plastizität und ihrer späteren molekularen Mechanismen zu studieren. Darüber hinaus in Kombination mit dem Einsatz von gentechnisch veränderten Tieren 15,16 und die Entwicklung von Gentransfer-Techniken (Adenovirus 2 oder lentiviralen Transduktion, Einsatz von Nanopartikeln, etc), diese in vitro primäre Azinuszelltumoren Modell kannsehr nützlich sein, zu bestimmen, wie verschiedene genetische Störungen der Regulierung der Azinuszelltumoren Differenzierung oder Entdifferenzierung beeinflussen und sollte ein besseres Verständnis der molekularen Vorgänge verantwortlich für das Auftreten von Pankreatitis, Krebsvorstufen und Veränderungen in Zellplastizität bieten. Isolation von dispergierten acini ist der Ansatz, die wir in unserem Labor zu Kultur Azinuszellen des Pankreas. Wir beschreiben und diskutieren die Methode verwendet. Es handelt enzymatische Dissoziation von Pankreasgewebe (mit einer bakteriellen Collagenase) gekoppelt mechanisches Aufbrechen ohne Dissoziation Azinuszellen. Während die meisten Protokolle Kultivieren der Acini, beinhalten entweder in Suspension oder auf speziell behandeltem Kunststoff Substraten wachsen wir sie in Suspension nur kurz (24 Std.), Säen sie anschließend auf einer Matrix Gerüste, wenn längerer Zellkultur erforderlich. Dieses Protokoll ermöglicht eine schnelle Trennung (in weniger als 1 Stunde) des Pankreas verteilt acini, nachhaltige für mehr als eine Woche in der Kultur. Es ermöglicht Isolierung von mehr als 20 x 10 6 Zellen pro Maus acinar Bauchspeicheldrüse. Die Einfachheit ist es möglich, unabhängig verarbeiten zu 10 Pankreas sind. Durch die Beibehaltung des intra-und interzellulären Architektur der Acini und damit die acinar Phänotyp der isolierten primären Zellen, stellt dieses Modell ein System der Wahl für die Untersuchung von Mechanismen transdifferentiation, wie alle anderen exokrinen derzeit verfügbaren Modelle von Tumoren der Bauchspeicheldrüse Anzeige von vielen genetischen abgeleitet Änderungen, die zu zellulärer Transformation.
