De toenemende vraag naar eiwit gebaseerde geneesmiddelen wereldwijd vereisen nieuwe productieplatforms die robuust, schaalbaar, lage kosten en een kluisje. Planten hebben aangetoond dat een van de meest veelbelovende alternatieve productiesystemen voor farmaceutische productie eiwit. In de afgelopen jaren is de ontwikkeling van deconstructed virus gebaseerde vectoren transiënte expressie van eiwitten in planten, die sterk verbetert de snelheid en opbrengst van plantaardige expressiesystemen 2,10 ingeschakeld...

Sinds 1970 zijn planten onderzocht als alternatieven voor zoogdieren, insecten en bacteriële celculturen voor de commerciële productie van recombinant proteïnen en therapeutische eiwitten 1. Plant-gebaseerde systemen voor de expressie van biofarmaceutische producten hebben aangetoond belofte in de afgelopen jaren een aantal nieuwe behandelingen voor ziekten, zoals de ziekte van Gaucher 2, en aviaire influenza H5N1 3, succesvol zijn gebleken in klinische onderzoeken. De ontwikkeling van competente mechanismen voor recombinante eiwitexpressie in planten in de decennia aangezien deze eerste experimenten heeft het potentieel voor plantaardige systemen voor het huidige paradigma van eiwitproductie aanpassen voor drie primaire redenen. Ten eerste is er een opmerkelijke afname in kosten bij zoogdieren, insecten en bacteriële bioreactoren vereisen aanzienlijke opstartkosten, duur groeimedia en gecompliceerde processen voor stroomafwaartse zuivering 4. De oprichting van stabiele transgene plantlijnen laat hen ook om de schaalbaarheid van andere expressie systemen overtreffen als eiwit expressie planten kunnen worden geteeld en geoogst op een agrarische schaal 5. Ten tweede, plantaardige expressiesystemen het risico op het overdragen van een mens of dier pathogeen het eiwit tot expressie gastheer voor mensen te verminderen, waaruit superioriteit openbare veiligheid 6. Tenslotte planten gebruik van een eukaryoot endomembranesysteem dat vergelijkbaar zoogdiercellen, waardoor goede post-translationele modificatie van eiwitten zoals glycosylering en de montage van meerdere-subeenheideiwitten 7. Dit vermogen geeft plantaardige systemen voordat deze basis prokaryotische systemen, zoals bacteriën, aangezien een groter aantal farmaceutische recombinante eiwitten, waaronder monoklonale antilichamen (mAbs) hebben een ingewikkeldere structuur en vereisen uitgebreide posttranslationele modificaties of assembly 8. Er zijn twee belangrijke passendpijn tot de uiting van recombinante eiwitten in planten. De eerste is de ontwikkeling van een transgene lijn stabiel, waarbij DNA dat codeert voor het doeleiwit wordt gekloneerd in een expressie cassette en ingevoerd om de nucleaire en chloroplastgenomen bedoeld. Daarbij, het vreemde DNA wordt erfelijk door opvolgende generaties en zorgt voor enorm verbeterde schaalbaarheid, veel verder dan die van andere systemen meningsuiting 1. Introductie van exogeen DNA aan het nucleaire genoom wordt gewoonlijk bereikt door Agrobacterium tumefaciens infectie van plantenweefsel of, minder vaak, door microprojectielbombardement van het weefsel 9. Plantenhormonen worden vervolgens gebruikt om differentiatie en groei van transgene plantenweefsel zoals wortels en bladeren induceren. Transformatie van de chloroplast genoom kan niet worden bereikt met A. tumefaciens, maar vertrouwt volledig op goud of wolfraam deeltjes bekleed met DNA afgevuurd ballistisch in plantencellen. De tweede methode van het uiten van recombinatient in planten is door tijdelijke expressie 10. In dit scenario worden virus afgeleide vectoren herbergen het gen van belang geleverd via A. tumefaciens tot volledig ontwikkelde planten door middel van een proces genaamd agroinfiltration. In plaats van integratie in het plantengenoom, wordt de geleverde genconstruct dan beginnen direct de tijdelijke productie van het gewenste eiwit, die kunnen worden geoogst en geïsoleerd na een korte incubatieperiode. Transiënte genexpressie biedt het voordeel van een grotere totale eiwit accumulatie en een verbeterde tijd eiwitproductie, zoals planten direct ongeveer 1-2 weken na oogsten agroinfiltration 11 zal zijn. Dit is aanzienlijk sneller dan de processen van productie, selectie, en de bevestiging van stabiele transgene plant lijnen, die enkele maanden kan duren tot een jaar. Dit is echter ook de beperking van het tijdelijke expressiesysteem, omdat het niet zal opleveren genetisch stabiele plantaardige lines die kunnen worden gebruikt om een ​​zaadbank voor grootschalige commerciële productie genereren. Desondanks zijn methoden ontwikkeld om grootschalige tijdelijke expressie verbeteren. Hier laten we zien een methode van generatie van voorbijgaande proteïne-expressie Nicotiana benthamiana planten met behulp gedeconstrueerd virale vectoren door A. geleverd tumefaciens. Twee belangrijke methoden ontwikkeld voor de levering van A. tumefaciens in plantenweefsel: bench schaal infiltratie via een injectiespuit en grootschalige infiltratie via de vacuümkamer. Beide protocollen worden beschreven met N. benthamiana, die nauw verwant is aan de gemeenschappelijke tabaksplant, zoals de waardplant voor transiënte expressie van twee fluorescerende eiwitten: het groen fluorescerend eiwit (GFP) van kwallen Aequorea Victoria en de rode fluorescente eiwit uit Discosoma koraal (DsRed) 12,13. N. benthamiana is de meest voorkomende waardplant voorrecombinant eiwit omdat het vatbaar is voor genetische transformatie kunnen grote hoeveelheden biomassa snel leveren en is een vruchtbare zaad producent opschaling productie 14. Een ander voordeel van N. benthamiana als gastheren voor eiwitexpressie is de beschikbaarheid van een verscheidenheid van expressievectoren 2,5. In deze studie, twee deconstructed virale vectoren, op basis van een tabak (TMV) RNA replicon systeem (MagnICON vectoren) en de andere zijn afgeleid van de boon yellow dwarf virus (BeYDV) DNA replicon systeem (geminiviral vectoren) 4,11, 15-18, worden gebruikt om het GFP en DsRed gen dragen en zal ze in N. benthamiana cellen via A. tumefaciens. Drie DNA constructen zullen worden gebruikt voor GFP en DsRed MagnICON expressie vectoren. Zij omvatten de 5 &#39;module (pICH15879) bevattende de promoter en andere genetische elementen voor het aandrijven van de expressie van het doelwitgen, het 3&#39;module die het gen van belang (Pich-GFP of Pich-DsRed) en het integrase module (pICH14011) codeert voor een enzym dat integreert de 5 &#39;en 3&#39; modules samen bij expressie 8,15. Drie DNA-constructen zijn ook nodig voor expressie met geminiviral vectoren. Naast de vectoren die replicon van het doelgen (pBYGFP of pBYDsRed), wordt een vector die codeert voor het replicatie-eiwit (pREP110) vereist voor de amplificatie van de doelwit replicon 11,14,16. efficient agroinfiltration of plants for high-level transient expression of recombinant proteins

Zoogdiercelculturen is het belangrijkste platform voor de commerciële productie van humane vaccins en therapeutische eiwitten. Het kan echter niet voldoen aan de toenemende wereldwijde vraag naar geneesmiddelen vanwege de beperkte schaalbaarheid en hoge kosten. Planten hebben aangetoond dat een van de meest veelbelovende alternatieve farmaceutische productieplatforms die robuust, schaalbaar, goedkoop en veilig. De recente ontwikkeling van virus-gebaseerde vectoren is toegestaan ​​snelle en hoog-niveau transiënte expressie van recombinant eiwitten in planten. Verder optimaliseren van de bruikbaarheid van het tijdelijke expressiesysteem, demonstreren we een eenvoudig, efficiënt en schaalbaar methodologie om target-gen dat Agrobacterium introduceren in plantenweefsel in deze studie. Onze resultaten geven aan dat zowel agroinfiltration spuit en vacuüm methoden hebben geleid tot de efficiënte introductie van Agrobacterium in bladeren en levenskrachtige twee fluorescerende eiwitten, GFP en DsRed. VoortsWe tonen de unieke voordelen van beide methoden. Spuit infiltratie is eenvoudig en dure apparatuur hoeft niet. Ook kan de flexibiliteit om ofwel infiltreren gehele verlaten met een doelgen, of genen van meerdere doelen voeren op een blad. Zo kan het worden gebruikt voor laboratoriumonderzoek expressie van recombinante eiwitten en voor verschillende eiwitten of vectoren waarin voor yield of expressie kinetiek. De eenvoud van de spuit infiltratie suggereert ook haar nut in de middelbare school en universiteit onderwijs voor het vak van de biotechnologie. Daarentegen, vacuüm infiltratie meer robuust en opgeschaald voor commerciële vervaardiging van farmaceutische eiwitten. Het biedt ook het voordeel van het kunnen plantensoorten die niet vatbaar is voor spuit infiltratie zoals sla en Arabidopsis zijn agroinfiltrate. Kortom, de combinatie van spuit en vacuüm agroinfiltration biedt onderzoekers en docenten een eenvoudige, efficiënte en robuustemethodologie voor transiënte eiwitexpressie. Het zal veel gemakkelijker de ontwikkeling van farmaceutische eiwitten en bevorderen van wetenschappelijk onderwijs.
