Leucine rich repeat kinasen 1 en 2 (LRRK1 en LRRK2) zijn multidomein paralogen die een soortgelijke domeinnaam organisatie delen. Beide eiwitten coderen een GTPase sequentie verwant aan de familie van Ras GTPases (Ras van complexe eiwitten, of ROC) en een C-terminale domein van ROC (COR), effectief classificeren beide eiwitten aan de ROCO eiwitfamilie 5,6. N-terminus van de ROC-COR domein tandem, beide eiwitten coderen een leucine-rich repeat domein en een ankyrine-achtige domein, terwijl slechts LRRK2 codeert extra armadillo domein 6-8. C-terminus van ROC-COR, beide eiwitten delen een serine-threonine kinase domein terwijl slechts LRRK2 codeert voor een WD40 domein in het C-terminale gebied 8. De precieze cellulaire rol van LRRK2 LRRK1 en moeten nog worden opgehelderd, maar LRRK1 is betrokken bij tyrosine kinase receptor signalisatie 1,2, terwijl genetische gegevens wijzen op een rol voor LRRK2 in de pathogenese van de ziekte van Parkinson 3,4. <pclass = &quot;jove_content&quot;&gt; De fosforylering van eiwitten is een gemeenschappelijk mechanisme in cellen. Leucine rich repeat kinases 1 en 2 (LRRK1 en LRRK2) zijn paralogen die eenzelfde domein organisatie delen, waaronder een serine-threonine kinase domein, een Ras van complexe eiwitten domein (ROC), een C-terminale domein van ROC (COR), en leucine-rijke en ankyrine-achtige herhalingen in de N-terminus. De precieze cellulaire rol van LRRK2 LRRK1 en moeten nog worden opgehelderd, maar LRRK1 is betrokken bij tyrosine kinase receptor signalisatie 1,2, terwijl LRRK2 is betrokken bij de pathogenese van de ziekte van Parkinson 3,4. In dit rapport presenteren we een protocol bij de LRRK1 en LRRK2 eiwitten te labelen in cellen met 32p orthofosfaat, waardoor een middel om de algemene fosforylering niveaus van deze 2 eiwitten in cellen te meten. Kortom, affiniteit gelabeld LRRK eiwitten worden uitgedrukt in HEK293T cellen die zijn blootgesteld aan medium met 32P-orthofosfaat. De 32P-orthofosfaat wordt opgenomen door de cellen na enkeleuur incubatie en alle moleculen in de cel met fosfaten daardoor radioactief gelabeld. Via de affiniteit tag (3xflag) de LRRK eiwitten worden geïsoleerd van andere cellulaire componenten door immunoprecipitatie. Immunoprecipitaten worden dan gescheiden via SDS-PAGE, geblot op PVDF membranen en analyse van de opgenomen fosfaten wordt uitgevoerd door autoradiografie (32 P-signaal) en Western detectie (eiwit-signaal) van de eiwitten op de blots. Het protocol kan gemakkelijk worden aangepast aan fosforylering van andere eiwitten die tot expressie worden gebracht in cellen en geïsoleerd door immunoprecipitatie controleren.
