Leucin rika upprepnings kinaser 1 och 2 (LRRK1 och LRRK2) är multidomain paraloger som delar en liknande organisation domän. Båda proteinerna koda ett GTPas-sekvens besläktad med Ras-familjen av GTPases (RAS av komplexa proteiner, eller ROC) såväl som en C-terminal av ROC-domänen (COR), vilket effektivt klassificera båda proteinerna till ROCO proteinfamiljen 5,6. N-terminalen av domänen tandem ROC-COR, båda proteinerna koda en leucinrik upprepningsdomän samt en ankyrin-liknande domän, medan endast LRRK2 kodar en extra bält Domein 6-8. C-terminal av ROC-COR, båda proteinerna delar ett serin-treonin-kinas-domänen medan endast LRRK2 kodar ett WD40 domänen i den C-terminala regionen 8. De exakta cellulära roller LRRK1 och LRRK2 har ännu inte klarlagd, men LRRK1 har varit inblandad i tyrosinkinasreceptor signalering 1,2, medan den genetiska bevisen pekar på en roll för LRRK2 i patogenesen av Parkinsons sjukdom 3,4. <pclass = &quot;jove_content&quot;&gt; Den fosforylering av proteiner är en gemensam rättslig mekanism i cellerna.
